Study of kinetics and mechanism of cholinesterases inhibition by benzothiazoles

Abstract:

Inhibitory cholinesteras jsou v současnosti prakticky jediné účinné látky, které zmírňují důsledky Alzheimerovy choroby i četných dalších onemocnění souvisejících s cholinergním deficitem. Práce je zaměřena na hledání nových inhibitorů cholinesteras s převahou působení v mozkové tkáni. Byla testována inhibiční schopnost dvanácti nových inhibitorů na bázi benzothiazolů dvěma na sobě nezávislými in vitro metodami (Ellmanovou metodou a pH-stat metodou), dále byl ověřen kinetický model neinhibovaných hydrolys. Pro posouzení účinnosti inhibitorů byl stanoven index IC50. Jako substrát pro pH-stat metodu byl užit acetylcholin, v případě Ellmanovy metody poté acetylthiocholin, použitými enzymovými preparáty pro obě zmíněné metody pak byly acetylcholinesterasa a butyrylcholinesterasa. Pro posouzení prostupnosti studovaných látek skrz hematoencefalickou bariéru byl změřen rozdělovací koeficient KOW. Nejperspektivnějšími látkami využitelnými pro klinickou praxi se jeví na základě vysoké inhibiční účinnosti a dobré rozpustnosti v nepolárních roztocích zejména inhibitory (S)-1-((L-leucinethyl)-1-(6-fluorobenzo[d]thiazol-2-yl)butylkarbamoyl)-2-methylpropylkarbamát a (S)-1-((L-phenylalaninethyl)-1-(6-fluorobenzo[d]thiazol-2-yl) propylkarbamoyl)-2-methylpropylkarbamát.

Abstract:

Nowadays, cholinesterases inhibitors are practically the only effective substances mitigating the impacts of Alzheimer's disease as well as of many other diseases related to cholinergic deficit. This piece of work focuses on the search for new cholinesterases inhibitors that effect mainly the brain tissue. The inhibition ability of twelve new inhibitors based on benzothiazole was tested by means of two mutually independent in vitro methods (the Ellman's method and the pH-stat method), the kinetic model of uninhibited hydrolysis was verified. To judge the effectiveness of the inhibitors the IC50 index was determined. The pH-stat method used acetylcholine as its substrate, the Ellman's method used acetylthiocholine. Both of the aforementioned methods used acetylcholinesterase and butyrylcholinesterase as its enzymatic preparations. To judge the permeability of the researched substances through the blood-brain barrier the partition coefficient Kow was determined. The most promising substances usable in clinical practice seem to be especially the (S)-1-((L-leucinethyl)-1-(6-fluorobenzo[d]thiazol-2-yl)butylkarbamoyl)-2-methyl propyl carbamate and (S)-1-((L-phenylalanin ethyl)-1-(6-fluorobenzo[d]thiazol-2-yl) propyl karbamoyl)-2-methyl propyl carbamate inhibitors, for their high inhibition efficiency and good solubility in nonpolar solutions.
