Structural and functional aspects of EGFR glycosylation in cancer

Abstract:

V této práci se zabývám aspekty glykosylace receptoru pro epidermální růstový faktor (EGFR) v kontextu maligní transformace buněk. EGFR je transmembránový glykoprotein, který se účastní proliferace, diferenciace a přežívání buněk, hraje důležitou roli v regulaci buněčného cyklu a jeho nadměrná stimulace může přispívat ke vzniku a šíření nádoru. Je proto často deregulován u mnoha typů karcinomů a je cílem moderních terapeutických látek, jako jsou monoklonální protilátky nebo malé molekulární inhibitory. Glykosylace patří mezi nejčastější posttranslační modifikace proteinů a má vliv na mnoho důležitých vlastností proteinů, jako je např. jejich konformace, stabilita nebo funkce. Extracelulární doména EGFR obsahuje mnoho připojených N-glykanů, které hrají roli ve vazbě ligandu na receptor, dimerizaci receptoru, endocytóze a také v senzitivitě k cílené anti-EGFR terapii. Cílem této práce je shrnutí současných poznatků o struktuře a funkci EGFR, vlivu specifické glykosylace EGFR na přenos signálu, prediktivní potenciál nádorových onemocnění a citlivost k různým formám protinádorové terapie.

Abstract:

In this thesis I focus on various aspects of epidermal growth factor receptor (EGFR) glycosylation in the context of malignant transformation of cells. EGFR is a transmembrane glycoprotein, which participace on cell proliferation, differentiation and survival; it plays an important role in cell cycle regulation and its upregulation may contribute to tumour formation and proliferation. EGFR is often deregulated in many types of cancer and thus it is a promising target of novel therapeutic drugs such as monoclonal antibodies or small molecular inhibitors. Glycosylation is one of the most common posttranslational modification of proteins and may affect many important features of protein biology, e.g. conformation, stability or function. EGFR extracellular domain contains many attached N glycans, which play a crucial role in ligand binding, receptor dimerisation, endocytosis and also in the sensitivity to anti-EGFR therapy. The aim of this study is to summarize current knowledge about EGFR structure and function with emphasis on the effects of specific EGFR glycosylation in signal transduction. Next a predictive potencial of EGFR glycosylation in different cancers and the sensitivity to various forms of cancer therapy is discussed.
