The role of macrophages in the regulation of iron metabolism during the metamorphosis of Drosophila melanogaster

Abstract:

Macrophages play a crucial role in the immune system by engulfing pathogens, clearing cellular debris and regulating inflammation. Their ability to handle residual cell components is largely studied in mammals, where it contributes to the recycling of iron - a vital but potentially lethal element if not regulated properly. The regulation is managed through a wide range of conserved iron-associated genes. This suggests that similar mechanisms are preserved and required in the metamorphosis of Drosophila melanogaster, as the fly undergoes a full body remodelling, especially of the fat body. The work aims to determine the efficiency of macrophage-specific RNA interference targeting selected genes involved in iron metabolism. Transgenic lines were generated and compared to a non-induced control group with quantitative PCR. Statistically significant gene knockdown was established, which can serve as a foundation for future studies investigating the impact of these genes on iron homeostasis.
