Research of endocrine disrupters and genotoxins in model aquatic ecosystem affected by the waste-water treatment plant

Abstract:

Hlavní náplní této práce bylo zhodnocení vlivu čistírny odpadních vod na říční tok Schussen v Německu. K tomuto účelu byly vzorky vod a sedimentů (odebírané v průběhu dvou let), podrobeny sadě in vitro biotestů, pokrývající nejčetnější formy specifického toxického působení znečišťujících látek. Jmenovitě byla stanovována genotoxicita SOS chromotestem, toxicita dioxinového typu na buněčné linii H4IIE.luc a endokrinní disrupce – estrogenita a antiestrogenita pomocí buněčných linií MVLN a hERα-HeLa-9903 a antiandrogenita na buněčné linii MDA-kb2. Extrakty vzorků vod nevykázaly žádné významné efekty vyjma testů genotoxicity, kdy se jako genotoxické projevily vzorky 3-6x zkoncentrované. Extrakty sedimentů se naopak ukázaly jako negenotoxické v testovaných koncentracích. Výsledky testů dioxinové aktivity uváděné v ekvivalentech TCDD byly v rozmezí 46-476 pg TCDD / g sušiny sedimentu. Výsledky testů estrogenní aktivity uváděné v ekvivalentech E2 byly v rozmezí <0,22 – 54 pg E2 / g sušiny sedimentu. Dále byl zaznamenán meziroční pokles všech sledovaných parametrů, avšak k potvrzení trendu je sledovaný časový úsek krátký.

Abstract:

The main object of this thesis was to evaluate effect of sewage treatment plant on river basin Schussen in Germany. For this purpose, water and sediment samples (collected during the two years’ time) have been used in sets of in vitro biotests, covering numerous forms of specific toxic impact of pollutants. In particular, genotoxicity has been specified by using the SOS chromotest, dioxin-like activity in H4IIE-luc bioassay and endocrine disruption – estrogenity and antiestrogenity using cell line MVLN and hERα-HeLa-9903, and finally anti-androgenity on MDA-kb2 cell line. Extractions of water samples haven’t shown any significant effects, except genotoxicity tests, where samples 3 to 6 time concentrated happened to be genotoxic. However, extracts of sediments haven’t appeared to be genotoxic in tested concentration. Results of dioxin activity bioassays, presented in TCDD equivalents, were in interval from 46 to 476pg TCDD/g of sediment dry matter. Results of bioassays for estrogenic activity, presented in E2 equivalents, were in interval from <0,22 to 54 pg E2 / g of sediment dry matter. Further on, I would like to notify year-on-year decrease of all of the followed parameters, although to confirm the trend, observing period of time was too short.
