Comprehensive strategy for decreasing a negative impact of Fusarium spp. toxicogenic fungi infection in cereals and their derivated products

Abstract:

Disertační práce se zabývá problematikou minimalizace negativního dopadu infekce působené patogeny rodu Fusarium na komoditu sladovnický ječmen. Za tímto účelem probíhaly v letech 2011 až 2013 maloparcelní experimenty s deseti přirozeně i uměle inokulovanými (F. culmorum) odrůdami (Aksamit, Bojos, Gladys, Kangoo, Malz, Prestige, Radegast, Sebastian, Tocada, Xanadu) ječmene jarního (Hordeum vulgare L.) na čtyřech lokalitách (Kroměříž, Libčany, Senice na Hané, Žabčice), ve kterých byl testován efekt aplikace (v růstových fázích BBCH 25–30, 39 a 65) kombinací fungicidních přípravků (Hutton®, Prosaro® 250 EC, Zantara®) s účinnými látkami (ú. l.) bixafen, prothiokonazol, spiroxamin a tebukonazol na výskyt patogenů v klasech, výnos (t/ha) zrna a obsahy (μg/kg) detekovatelných mykotoxinů v zrnu (DON – deoxynivalenol, D3G – deoxynivalenol-3-glukosid, ADONs – 3- a 15- acetyldeoxynivalenol, NIV – nivalenol, α-ZOL – α-zearalenol, β-ZOL – β- zearalenol, HT2 a T2 toxin, BEA – beauvericin, Enn – enniatiny A, A1, B, B1). Ke stanovení mykotoxinů byla použita metoda QuEChERS. Výskyt hub r. Fusarium v klasech byl nejčastěji spojen s odrůdami Gladys a Xanadu, naopak tomu bylo u odrůdy Malz. V podmínkách přirozené kontaminace převažovaly v zrnech nálezy F. poae, patogen ale na zvolená fungicidní ošetření nereagoval, ostatní fuzáriové patogeny ú. l. tebukonazol a prothiokonazol převážně potlačovaly. Všechna fungicidní ošetření působila jako výnos zrna „ochraňující“. Kromě běžných toxinů (NIV, DON, D3G, ADONs, HT2 a T2) se ve vysokých hladinách vyskytovaly v přirozeně kontaminovaných zrnech také „emerging“ mykotoxiny (EM – ENNs a BEA). Fungicidní aplikace snižovaly hladiny trichothecenů B i ZEA, neprokázaly ale účinnost při snižování obsahů NIV a EM. Nejvíce mykotoxinů akumulovaly v přirozených podmínkách Tocada (5735 μg/kg), Xanadu (3177), Prestige (3102) a Kangoo (2992), nejméně Radegast (1652) a Bojos (1699).

Abstract:

The thesis is focused on the issues associated with minimisation of the negative impact of infection caused by Fusarium genus pathogens to the malting barley commodity. Small-plot experiments using ten naturally contaminated and artificially inoculated (F. culmorum) spring barley (Hordeum vulgare L.) varieties (Aksamit, Bojos, Gladys, Kangoo, Malz, Prestige, Radegast, Sebastian, Tocada, Xanadu) were carried out for this purpose during three consecutive years 2011 – 2013 at four locations (Kroměříž, Libčany, Senice na Hané, Žabčice). The effect of application was studied (at growth stages BBCH 25–30, 39 and 65) for selected combinations of fungicide preparations (Hutton®, Prosaro® 250 EC, Zantara®; active substances: bixafen, prothioconazole, spiroxamine and tebuconazole) on the occurrence of pathogens in ears, yield of grain (t/ha) and the content of detectable mycotoxins (μg/kg) in grain (DON – deoxynivalenol, D3G – deoxynivalenol-3-glucoside, ADONs – 3- and 15- acetyldeoxynivalenol, NIV – nivalenol, α-ZOL – α-zearalenol, β-ZOL – β-zearalenol, HT2 and T2 toxins, BEA – beauvericin, ENN – enniatins A, A1, B and B1). The QuEChERS method was used for mycotoxin determination. The occurrence of Fusarium fungi in ears was most often observed in the varieties Gladys and Xanadu; the opposite case was the variety Malz. Under the conditions of natural contamination, F. poae was prevalent pathogen found in the grains; however, it did not respond to the selected fungicide treatments. The other Fusarium spp. pathogens were mostly suppressed by tebuconazole and prothioconazole fungicides. All fungicide treatments acted as "protecting" grain yield. In naturally contaminated grains, not only common toxins (NIV, DON, D3G, ADONs, HT2 and T2), but also so called “emerging mycotoxins” (EM – ENNs and BEA) were present at high concentrations. Fungicide applications were able to reduce the levels of trichothecenes B and ZEA, but were not so effective as for NIV and EM contents. The most naturally-infected varieties accumulated highest amounts of mycotoxins in following order: Tocada, Xanadu, Prestige and Kangoo (5735, 3177, 3102, and 2992 μg/kg, respectively), the lowest contamination was observed for Radegast and Bojos (1652 and 1699 μg/kg, respectively).
