Abstract:

Proteiny jsou častým cílem posttranslačních modifikací, které jim dávají nové vlastnosti. V této diplomové práci jsme se zabývali proteinem Dishevelled, který je jednou z podstatných komponent signalizace Wnt a který reguluje její průběh. Pomocí hmotnostní spektrometrie jsme analyzovali fosforylace proteinu hDVL3 indukované osmi asociovanými kinázami přičemž byly nalezeny desítky fosforylovaných aminokyselin. Hlavním výstupem této práce tedy je fosforylační „mapa“ proteinu Dvl, která určila místa specifická pro jednotlivé kinázy. Vliv fosforylovaných aminokyselin na průběh Wnt signalizace jsme se snažili ověřit také funkčními testy. Vybrané fosforylované seriny/threoniny byly mutovány na alanin a byl zkoumán důsledek těchto mutací hDVL3 na průběh Wnt/β-kateninové dráhy a na změnu elektroforetické migrace hDVL3, která je jedním ze znaků aktivované Wnt signalizace. Také jsme ověřili funkčnost fosfo specifických protilátek proti hDVL3, avšak pouze část z připravených protilátek se ukázala býti specifická. Informace v této práci obsaženy byly a dále budou využity pro další výzkum vlivu proteinu Dvl na Wnt signální dráhy.

Abstract:

Proteins are frequent targets of posttranslational modifications, which give them new properties. In this thesis we studied Dishevelled protein, essential component of Wnt signalling pathway that contributes to their regulation. We performed mass spectrometric analysis of hDVL3 phosphorylations induced by eight associated kinases, which revealed several dozens of phosphorylation sites. Main outcome of this thesis is therefore phosphorylation “map” of Dvl protein that attributed phosphorylation sites for individual kinases. We performed functional tests of influence of selected phosphorylations on Wnt signaling pathways. Selected phosphorylated serines/threonines were mutated to alanine and the impact of these mutations on Wnt/β-catenin signalling pathway and on the change of hDVL3 electrophoretic migration, one of the signs of Wnt pathway activation, was investigated. We also verified functionality of anti-phospho hDVL3 antibodies, but not all of them were found specific. Contents of this thesis were and will further be used for the analysis of influence of Dvl on the Wnt signaling pathways.
