Characterization of red clover genes specific for biotic stress

Abstract:

Jetel luční (Trifolium pratense L.) je významná pícnina se schopností fixovat vzdušný dusík díky symbióze s hlízkovitými bakteriemi a šlechtitelské snahy se proto ubírají jak směrem k identifikaci rostlinných genů podílejících se na fixaci, tak charakterizaci genů podílejících se na obraně rostliny vůči stresu. Účelem této diplomové práce bylo studium vybraných kandidátních genů biosyntetické dráhy vedoucí k syntéze fytoalexinů u T. pratense s využitím dat získaných sekvenováním jeho genomu a následná bioinformatická a experimentální analýza. Stěžejní části experimentálních analýz byly expresní analýzy s cílem ověřit expresi vybraných genů a zjistit, jestli existuje nějaká korelace mezi expresí těchto genů a fenotypovým znakem odolnosti vůči patogenu Erysiphe trifolii. Tyto analýzy pomůžou lépe pochopit vztah mezi expresí vybraných genů a jejich účinkem na fenotyp a zhodnotit jejich potenciál ve šlechtění rostlin.

Abstract:

Trifolium pratense L. is an important fodder crop with the ability to fix air nitrogen due to symbiosis with nitrogen-fixing bacteria, and breeders' efforts are directed both towards identification of plant genes involved in nitrogen fixation and characterization of genes involved in plant defense against stress. The aim of this diploma thesis was the study of selected candidate genes involved in biosynthetic pathway leading to synthesis of phytoalexins in T. pratense using data obtained by genome sequencing and subsequent bioinformatics and experimental analysis. The main part of the experimental analyzes were expression analyses to verify the expression of candidate genes and to determine whether there is a correlation between the expression of these genes and the phenotypic trait of resistance against the fungal pathogen Erysiphe trifolii. These analyses can help to better understand the relationship between expression of candidate genes and their effect on the phenotype and evaluate their potential in plant breeding.
