Abstract:

Karcinom ovaria patří mezi nejzhoubnější gynekologické novotvary. K jeho léčbě se využívá operační přístup následovaný adjuvantní chemoterapií na bázi platinových cytostatik a paklitaxelu. Navzdory dobré počáteční odpovědi na léčbu dochází u dvou třetin pacientek k relapsu a rozvoji rezistence. K překonání výše zmíněných nevýhod platinových léčiv jsou vyvíjena nová chemoterapeutika s rozdílnou aktivitou k různým typům nádorů, odlišným mechanismem účinku, vyšší selektivitou k nádorovým buňkám a tím i nižší toxicitou pro zdravé buňky organismu. Stále více pozornosti se soustřeďuje na vývoj a výzkum účinků organokovových látek nesoucí ve své struktuře jiný kov než je platina. Cílem této práce bylo zhodnotit cytotoxickou aktivitu nových organokovových látek - titanocenů a ferocenů jako potenciálních protinádorových sloučenin, objasnit mechanismus jejich působení a vyvinout způsob detekce těchto sloučenin v buněčném lyzátu. Z dosažených výsledků vyplynulo, že titanocen difluoridy ve srovnání s testovanými dichloridy jsou cytotoxicky účinnější. Dále bylo zjištěno, že sloučeniny obsahující pouze jeden cukerný ligand navázaný na jeden ze dvou cyklopentadienidových kruhů jsou méně toxické pro vybrané ovariální nádorové linie než sloučeniny, které mají sacharidovou skupinu na obou Cp kruzích. Na základě MTT dat bylo ke studiu mechanismu působení vybráno několik zástupců difluorovaných titanocenů. Bylo zjištěno, že titanocen difluoridy tdf1 a tdf2 narušují homeostázu endoplazmatického retikula a aktivují alternativní mechanismy buněčné smrti. Dále byla vyvinuta elektrochemická metoda detekce nového derivátu ferocenu, která umožňuje rozlišit μM koncentrace dané látky v lyzátu nádorových buněk.

Abstract:

Ovarian cancer belongs to the most malignant gynecologic cancers. The treatment involves surgery followed by adjuvant chemotherapy based on platinum and paclitaxel. Despite of good initial response among two thirds of patients disease returns and develops cisplatin resistence. To overcome these disadvantages new chemotherapeutics with different mechanism of action, higher selectivity and lower toxicity to healthy cells are developed. The ongoing research and development is focused on non-platinum compounds. The aim of this work was to evaluate cytotoxic activity of titanocene and ferrocene compounds as potential antitumor drugs, elucidate the mechanism of action of selected substances and find a method for the detection of these organometallic compounds in the cell lysate. Increased cytotoxic activity of fluorinated titanocenes in comparison with chlorinated ones has been observed. It has been demonstrated that titanocenes dihalogenides bearing two carbohydrate moieties (one on each Cp ring) show higher cytotoxicity than their counterparts with only one carbohydrate moiety. Based on MTT results several titanocen difluorides have been chosen to study their mechanism of action. Our results indicate that titanocene difluorides tdf1 and tdf2 disturb endoplasmic reticulum homeostasis and trigger autophagic mechanisms of cell death.Taken together our data suggest that replacement of chloride ligands by fluoride together with introduction of carbohydrate-derived substituents at the cyclopentadienide ligand is a promising strategy for the development of novel metallocene-based anticancer compounds. Moreover we developed quick and inexpensive electrochemical method for the determination of novel ferrocene derivative with cytotoxic properties in the low µM range directly in cancer cell lysates. This method could be used to determine the ability of newly synthesised compounds with putative anticancer properties to penetrate the cell membrane.
