Monitoring of milk and milk fat changes during heating by luminescence spectroscopy

Abstract:

Diplomová práce je věnována možnostem využití metody luminiscenční spektroskopie pro sledování změn mléka a mléčného tuku během záhřevu. Teoretická část práce je věnována popisu chemického složení mléka a mléčného tuku, změnám jejich chemického složení v průběhu tepelného záhřevu a obecné charakteristice luminiscenční spektroskopie. Praktická část je orientována na naměření luminiscenčních spekter mléčného tuku a mléka s různou tučností, zachycení a vyhodnocení jejich změn v závislosti na teplotě a délce zá-hřevu vzorků. U vzorků mléčného tuku byly vyhodnocovány píky s emisí při vlnové délce 410 nm a 525 nm s použitím excitační vlnové délky 315 nm. U sledovaných píků byla za-znamenána významná změna intenzity luminiscence v intervalu 120-140 °C, který odpoví-dá hodnotám kouřového bodu másla. U vzorků mléka byla interpretace výsledků ztížena jeho vícesložkovou strukturou poskytující široké překrývající se fluorescenční pásy. Byl hodnocen pík s emisí 345 nm při použití excitační vlnové délky 280 nm, který vykazoval s rostoucí teplotou pokles intenzity emise.

Abstract:

This thesis deals with the possibilities of using the luminescence spectroscopy method for monitoring of milk and milk fat changes during heating. The theoretical part of the thesis is a summary description of chemical composition of milk and milk fat, changes in their chemical composition during the thermal heating and also the general characteristics of luminescence spectroscopy. The practical part of the thesis is focused on the measurement of milk fat luminiscence in milk samples with different fat content; interception and evaluation of changes depending on the temperature and time of sample heating. For milk fat samples, peaks at 410 nm and 525 nm were observed using an excitation wavelength of 315 nm. For these specific peaks, a significant change in luminescence intensity was observed when the temperature of samples was within the range of 120 - 140°C, which corresponds to the smoke point of butter. But there was a difficulty in interpretation of the milk samples results, caused by milk multicomponent structure providing broad overlapping fluorescent bandwidths. Using an excitation wavelength of 280 nm, a specific 345 nm peak was evaluated; and it was found out, that increasing temperature causes emission intensity decrease.
