The major ions of intracellular and extracellular fluid

Abstract:

Tato bakalářská práce se zabývá hlavními ionty intracelulární a extracelulární tekutiny. Pozornost je zaměřena na chloridové ionty a na metody jejich stanovení. Cílem práce je zavedení coulometrické titrace do provozu OKB FNO. Teoretická část popisuje dělení celkové vody v lidském organismu, osmolalitu, systémy udržující rovnováhu vody a iontů v našem těle, a samotné ionty intracelulární a extracelulární tekutiny: sodný, chloridový, vápenatý, draselný a hořečnatý ion. Praktická část bakalářské práce se zabývá stanovením chloridových iontů v potu. Spolu s onemocněním cystické fibrózy jsou zde popsány dvě metody, merkurimetrická a coulometrická titrace. Cílem praktické části je zavedení metody coulometrické titrace do provozu za účelem nahrazení metody merkurimetrické titrace, která se již na OKB FNO používá. V rámci zavedení nové metody byla provedena verifikace. Ta ověřila, že požadavky na metodu byly splněny a coulometrickou titraci lze zavést do provozu FNO.

Abstract:

This bachelor´s thesis deals with the major ions of intracellular and extracellular fluid. The attention is concentrated on the chlorides and methods of their determination. The aim of the bachelor´s thesis is implementation of method coulometric titration into the operation of the Institute of Clinical Biochemistry in the University Hospital Ostrava. The theoretical part of the bachelor´s thesis describes the division of total water in organism, osmolality, systems which keep balance of water and ions in our body, and the major ions of intracellular and extracellular fluid: sodium, chloride, calcium, potassium and magnesium. The practical part of the bachelor´s thesis deals with determination of chlorides in sweat. Together with the disease cystic fibrosis there are described two methods: mercurimetric and coulometric titration. The aim of the practical part is implementation of method coulometric titration into the operation with the view of supplying method mercurimetric titration which is actually running in the Institute of Clinical Biochemistry in the University Hospital Ostrava. In terms of introduction of the new method was implemented a verification. The verification proved that the requirements were realized and coulometric titration can be used to the operation of the University Hospital Ostrava.
