The role of Ah receptor in regulation of functions of cells of connective tissues

Abstract:

Receptor pro aromatické uhlovodíky (AhR) je významný transkripční faktor uplatňující se v regulaci metabolismu toxických látek. Předložená bakalářská práce se zaměřuje na funkci tohoto transkripčního faktoru v podpůrné pojivové tkáni, ve chrupavce a kostech, s ohledem na přítomné buňky mezenchymálního původu. Aktivace AhR v těchto tkáních může vést k narušení signálních drah zapojených do vývoje a správné funkce kostí a chrupavky, co přispívá ke vzniku kostních malformací a poruch podpůrné pojivové tkáně. Působení toxických ligandů AhR se projevuje sníženou diferenciací osteoblastů a zvýšenou tvorbou osteoklastů, co se následně projevuje zvýšenou resorpcí kostí. Aktivace AhR ve chrupavce má za následek vznik volných kyslíkových radikálů a následnou apoptózu chondrocytů. Z dostupné literatury je zřejmé, že aktivace AhR může ovlivnit funkce buněk kostí i chrupavky, a mechanismům působení toxických environmentálních ligandů AhR v buňkách pojivových tkání by tak měla být věnována zvýšená pozornost.

Abstract:

Aryl hydrocarbon receptor (AhR) is an important transcription factor involved in regulation of metabolism of toxic compounds. Submitted bachelor thesis focuses on the role of this transcription factor in supportive connective tissue, cartilage and bone, with respect to the cells of mesenchymal origin. Activation of AhR in this tissue may result in disruption of signalling pathways involved in development and function of cartilage and bones, which then contributes to the formation of bone malformations and illnesses of supportive connective tissue. The effects of toxic AhR ligands may lead to a decreased differentiation of osteoblasts and increased formation of osteoclasts, which consequently increases bone resorption. Activation of AhR in the cartilage results in production of reactive oxygen species followed by apoptosis of chondrocytes. The available literature suggests that activation of AhR may affect functions of bone and cartilage cells; therefore, an increased attention should be paid to the mechanisms of action of toxic environmental AhR ligands within connective tissues.
