Influence of ions on binding properties of lectines
Abstract:
V této práci jsou předmětem studia proteiny PA-IIL pocházející z bakterie Pseudomonas aeruginosa a BC2L-A pocházející z bakterie Burkholderia cenopacia, v jejichž vazebných místech se nacházejí dva vápenaté ionty podílející se na vazbě se sacharidem. Cílem práce bylo odebrat tyto vápenaté ionty z vazebného místa lektinů, nahradit je jinými dvojmocnými kovovými ionty a poté měřit jejich aktivitu a vazebné …more
Abstract:
The object of the study are proteins PA-IIL from bacteria Pseudomonas aeruginosa and BC2L-A from bacteria Burkholderia cenopacia, in whose binding site there are two calcium ions, that mediate binding between the lectin and a sacharide. The goal of this work is to remove these two calcium ions from the protein's binding site, replace them with other metal ions and then measure their activity and binding …more
