Antimicrobial Activity of Essential Oils on Bacteria Producing Biogenic Amines

Abstract:

V diplomové práci byl sledován vliv vybraných esenciálních olejů na růst bakteriálních kmenů produkujících biogenní aminy. Teoretická část klasifikuje použité bakteriální kmeny, chemické složení esenciálních olejů a specifikace sledovaných biogenních aminů. Praktická část byla věnována antibakteriálním účinkům použitých esenciálních olejů. Z výsledků je patrné, že minimální inhibiční koncentrace se u testovaných olejů pohybovala v rozmezí 250 - 1500 mg/l. Dále byl sledován vliv esenciálních olejů na produkci biogenních aminů fenylethylaminu, putrescinu, kadaverinu, histaminu a tyraminu u vybraných dekarboxyláza-pozitivních bakterií. Výsledky poukazují na dosažení minimální inhibiční koncentrace (250 mg/l) u rodu Lacctococcus při aplikaci esenciálního oleje z oregana. U bakteriálního kmene Enterococcus durans CCDM 53 bylo zaznamenáno největší snížení produkce biogenního aminu kadaverinu až o 80 %. Použitá koncentrace esenciálního oleje byla 1000 mg/l . Celkově gram-pozitivní bakterie byly vnímavější na aplikaci testovaných esenciálních olejů. Snížená produkce biogenních aminů u gram-negativních bakterií byla zjištěna u kmene Proteus mirabilis CCM 7188, kdy bylo zjištěno přibližné snížení o 20 % působením esenciálních olejů z tymiánu, rozmarýnu a lemongras, každý v koncentraci 500 mg/l. Esenciální olej z tymiánu měl při koncentraci 1000 mg/l vliv na snížení produkce biogenního aminu kadaverinu u bakteriálního kmene Salmonella enterica subsp. enterica ser. Enteritidis CCM 4420.

Abstract:

In this diploma thesis was observed the effect of some essential oils on the growth of bacterial strains which produce biogenic amines. The theoretical part classifies the bacterial strains, chemical compound of essential oils and the specification of observed biogenic amines. The practical part was focused on antibacterial effect of the essential oils. The results show that minimal inhibitory concentration of used oils was in the range 250 - 1500 mg/l. Next was observed the effect of essential oils on the production of biogenic amines phenylethylamine, putrescine, cadaverine, histamine and tyramine at decarboxylase-positive bacteria. The results show that achievement of a minimum inhibitory concentration (250 mg/l) on Lacctococcus in the application of oregano essential oil. The highest decrease in production of biogenic amine cadaverine, up to 80%, was recorded at the strain Enterococcus durans CCDM 53. The concentration of the oil used was 1000 mg/l. Overall gram-positive bacteria were more sensitive to the application of essential oils. The decrease in production of biogenic amines at gram-negative bacteria was recorded at the strain Proteus mirabilis CCM 7188, the decrease was approximately 20% with the usage of essential oils from thymus, rosemary and lemongrass, each in concentration 500 mg/l. The thymus essential oil in concentration 1000 mg/l influenced the decrease in production of biogenic amine cadaverine at the strain Salmonella enterica subsp.entericaser. Enteritidis CCM 4420.
