MHC genes in birds - comparison of model and wild-living species

Abstract:

Geny hlavního histokompatibilního systému (Mhc geny) jsou jednou ze základních složek imunitního systému obratlovců. Kódují proteiny na povrchu buněk, které vážou cizorodé antigeny a tyto jsou pak rozpoznávány T lymfocyty, čímž dojde k iniciaci imunitní reakce. Struktura Mhc genů je u různých druhů velmi variabilní a extrémní mezidruhové rozdíly jsou zejména u ptáků, což má samozřejmě dalekosáhlé evoluční a ekologické důsledky. Cílem této práce bylo provést rozsáhlou literární rešerši o struktuře, diverzitě a metodách analýzy Mhc genů u všech dosud studovaných druhů ptáků a osvojit si metodiku MHC analýzy u modelového druhu zebřičky pestré (Taeniopygia guttata). Do teoretické části byly zahrnuty druhy z celkem 11 řádů, včetně doposud nejlépe prostudovaných hrabavých a pěvců, u kterých existují nějaké informace o MHC komplexu. Popis genetických metod analýzy MHC byl zaměřen na nejčastěji využívané metody v současné době a blízké budoucnosti. Výsledky praktické části ukázaly u zebřiček nižší variabilitu Mhc genů než u jiných druhů pěvců. Bylo izolováno celkem 8 alel exonu 3 Mhc I nacházejících se nejméně na 2 lokusech. Následné analýzy ukázaly, že na sekvence působí silná pozitivní selekce a nachází se zde minimálně jedno místo rekombinace. Všechny sekvence tvoří ve fylogenetickém stromu s dalšími druhy pěvců samostatnou větev. V práci byl tak popsán vhodný marker pro popis Mhc I u zebřiček, který v budoucnu může sloužit pro další studie týkající se např. pohlavního výběru.

Abstract:

Major histocompatibility complex (Mhc) genes are one of the basic components of the immune system in vertebrates. They encode glycoproteins situated on the surface of cells, which bind nonself antigens. These complexes are subsequently recognised by T-lymphocytes, thus initiating the adaptive immune response. The structure of Mhc genes is very variable among species, which is particularly true in birds, and this fact may have important evolutionary and ecological consequences. The aim of this bachelor thesis was (1) to provide extensive review concerning structure, diversity and methods of Mhc genes analysis in all bird species studied to date, and (2) to acquire the practical methodology of Mhc analysis in a model species, the zebra finch (Taeniopygia guttata). The first theoretical part concerns species from 11 orders, including the most studied groups of galliform and passerine birds. The second theoretical part reviews the current laboratory techniques for analyses of Mhc genes. The results of the practical part showed lower variability of Mhc genes in zebra finches than in other passerine species. Within three individuals, eight alleles of exon 3 Mhc Class I were identified on at least two loci. Subsequent sequence analyses showed that there is strong positive selection mainly on antigen-binding sites. All zebra finch sequences formed a separate cluster in phylogenetic tree distinct from all other passerine sequences. The described Mhc Class I marker in zebra finches might be used in future studies of this model species, e.g. in mate choice research.
