Understanding molecular pathogenesis of diseases through advanced DNA technologies

Abstract:

Hledání molekulární patogeneze onemocnění je složitý a multioborový proces, ve kterém můžou moderní technologie DNA diagnostiky znamenat přínos nových informací. Tato práce proto v úvodní části přináší přehled vybraných základních a na ně navazujících pokročilých metod, které se obvykle používají ke studiu molekulární podstaty vzniku onemocnění. V mé dizertační práci se zaměřuji na tři vybrané multifaktoriální onemocnění – schizofrenii, Alzheimerovu nemoc a věkem podmíněnou makulární degeneraci. Všechny tyto nemoci nejsou doteď zcela prozkoumány, a to právě kvůli jejich velké variabilitě a obrovskému množství genetických i environmentálních faktorů, které je ovlivňují. V této práci shrnuji informace, které jsou k daným onemocněním aktuálně dostupné a dále se věnuji výzkumům, na kterých jsem se mohla podílet. U schizofrenie jsme potvrdili předpokládanou souvislost s genem pro inzulin degradující enzym (který je úzce spjatý s diabetem), založenou na častém výskytu diabetu 2. typu u pacientů se schizofrenií. Alzheimerova nemoc představuje riziko pro stále větší část populace, a proto se jejímu výzkumu věnuje velká pozornost. Našemu týmu se podařilo na české populaci popsat vliv genů pro ChAT a CD36 na patogenezi této nemoci. Gen pro CD36 souvisí také s věkem podmíněnou makulární degenerací, konkrétně s hladinami nitroočního tlaku během léčby nemoci. V neposlední řadě se nám podařilo zjistit asociaci mezi polymorfismy dalších čtyř genů (ARMS2, CFH, C3 a HTRA1) a vlhkou formou tohoto onemocnění oka.

Abstract:

Finding of molecular pathogenesis pathways for different diseases is a complicated process, in which DNA diagnostic technologies are a valuable source of new information. This thesis brings an overview of basic and related advanced methods, which are commonly used for the study of molecular pathogenesis of diseases. The centre of my thesis are three chosen multifactorial diseases – schizophrenia, Alzheimer´s disease and Age-related Macular Degeneration. These diseases are very hard to study because of their huge variability and the amount of genetic and environmental factors which can influence them. In my work, I summarize the information, which are currently available and describe the research I took part on. Frequent comorbidity between schizophrenia and type 2 diabetes mellitus brought us to assumption, there could be a relationship between insulin-degrading enzyme gene (which is closely related to T2DM) and schizophrenia, which we managed to prove. Alzheimer´s disease represents a risk for huge part of the population and therefore a huge attention is given to the research of this disease. Our team managed to describe the influence of ChAT and CD36 genes on Alzheimer´s disease pathogenesis. The CD36 gene also takes part on age-related macular degeneration pathogenesis, most of all on intraocular pressure during the treatment. We further found an association between four other genes (ARMS2, CFH, C3 and HTRA1) and the wet form of AMD.
