Complexity in behavioural organization: a novel approach to assessing parasite infection

Abstract:

Ve volné přírodě stejně jako v lidské péči mohou paraziti a parazitózy znamenat vážnou hrozbu pro zdraví zvířat. Symptomy parazitárních infekcí nejsou vždy viditelné, především pak u volně žijících zvířat, u kterých je obtížné pozorovat klinické příznaky. Posun k velkým komerčním chovům u hospodářských zvířat vede k nutnosti zavedení efektivního monitoringu zdravotního stavu zvířat. Již po dlouhou dobu byly změny v chování zvířat považovány za ukazatele stavu welfare zvířat, ale až nedávné studie prokázaly, že i zanedbatelné změny v chování mohou být spojeny se stresem, onemocněními nebo jinými ohrožujícími faktory. Implementace nových analytických nástrojů, které jsou schopny zaznamenat změny v organizaci chování, otevírá slibné možnosti neinvazivní detekce různých zdravotních problémů. Tato práce si klade za cíl zjistit, zdali míra komplexity chování může poskytnout informace o parazitárních onemocněních u volně žijících i hospodářských zvířat. Konkrétně jsem pomocí fraktálních analýz hodnotila potencionální vliv strongylidních hlístic na komplexitu chování. Volně žijící šimpanzi a domácí ovce byli vybráni jako reprezentativní modelové druhy, na nichž bylo zkoumáno použití fraktálních metod pro hodnocení parazitárních infekcí, a to v obou typech prostředí – ve volné přírodě a ve farmovém chovu. U divokých šimpanzů v Ugandě byla data o chování získána přímými pozorováními. Intenzita infekce strongylidních hlístic byla stanovena pomocí kvantifikace počtu vajíček v gramu sedimentu trusu, zatímco komplexita u sekvencí potravního chování byla hodnocena za pomoci tzv. Detrended Fluctuation Analysis (DFA), statistické metody určené k hodnocení zprostředkovaných korelací neboli „paměti“ u sekvenčních dat. Vztah mezi změnami v chování a intenzitou strongylidních infekcí byl následně testován vícerozměrnými statistickými metodami. Dále jsem zkoumala možnou variabilitu u více konvenčních parametrů popisujících chování, jako jsou celková délka, frekvence a průměrná délka ve vztahu k intenzitě infekce. U domácích ovcí byly údaje o aktivitě získány pomocí akcelometrických zapisovačů dat, které kontinuálně zaznamenávaly pohyby jejich těl ve třech osách. Abychom se vyhnuli zavádějícím výsledkům, které mohou vzniknout při spoléhání se na jeden typ fraktální analýzy, byly použity metody box-counting a upravený periodogram výsledných sérií akceleračního času a navíc dvě varianty DFA. Abychom otestovali vliv parazitární infekce na organizaci chování, porovnali jsme vztah mezi parasitární infekcí a komplexitou u vzorců aktivity u přirozeně infikovaných a experimentálně odčervených ovcí. Testovány byly i možné změny u více tradičních parametrů chování ve vztahu k infekčnímu statusu ovcí. U volně žijících i domácích zvířecích modelů mé celkové výsledky naznačují významnou variabilitu v časové organizaci chování ve vztahu k infekci strongylidními hlísticemi. U šimpanzů se projevily komplexnější sekvence potravního chování s nárůstem intenzity infekce a odčervené ovce vykazovaly vyšší komplexitu v jejich aktivitě v porovnání s infikovanými ovcemi. Zajímavé je, že v případě použití více konvenčních analýz chování nebyl prokázán žádný vliv parazitární infekce. Tyto výsledky podporují narůstající důkazy, že fraktální analýzy mohou být použity jako citlivá metoda pro monitorování welfare zvířat a mohly by podpořit budoucí využití analýz komplexity chování za účelem diagnostiky a monitorování trajektorie parazitárních infekcí. V dodatečné studii byl zkoumán možný vliv strongylidních infekcí na přítomnost DNA Plasmodium spp. (malárie) v trusu lidoopů, avšak nebyla

nebyla nalezena žádná jasná souvislost mezi těmito dvěma infekcemi a tudíž krvácení způsobené určitými strongylidními hlísticemi nemůže vysvětlit přítomnost malarické DNA v trusu lidoopů.

Abstract:

In the wild as well as in captivity, parasites and parasitic diseases can pose a serious threat to animal health. Symptoms of parasite infections are not always visible and even when clinical signs occur, it can be difficult to observe them in wild animals living in their natural environment. On farms, the move toward large commercial settings calls for the development of effective health monitoring. Behavioural changes have long been used to assess animal welfare, but more recently even more subtle variations in the organization of behaviour have been linked to stress, diseases or other challenging conditions. The emergence of innovative analytical tools that measure behavioural organization now offers promising opportunities to non-invasively detect various health impairments. This thesis aims at testing whether complexity measures can provide indications about the state of parasitic infections of both wild and domestic animals. More specifically, I assessed the potential effect of strongylid nematode parasites on behavioural complexity using fractal analyses. Wild chimpanzees and domestic sheep were chosen as representative models to explore the validity of using fractal methods to assess parasite infection, in both contexts of wildlife and on-farm health monitoring. In these two species, the temporal structure of behavioural sequences was examined by applying fractal analysis. In wild chimpanzees in Uganda, behavioural time series were collected by direct observations. The intensity of strongylid infection was estimated by quantifying the number of eggs per gram of faecal sediment, while complexity in feeding sequences was estimated using Detrended Fluctuation Analysis (DFA), a statistical method designed to assess long-range correlations or “memory” in sequential data. The relationship between changes in behavioural complexity and intensity of strongylid infections was then tested using multivariate statistics. Additionally, I investigated for potential variation in more conventional statistical measures of behaviour such as total duration, frequency and mean duration in relation to intensity of infection. In domestic sheep, activity sequences were obtained via accelerometer loggers that continuously recorded their body movements along three axes. To avoid misleading results that can arise when relying on a single form of fractal analysis, the box-counting method and a modified periodogram method were applied to the resultant acceleration time series in addition to two variants of DFA. In order to test for the effect of parasite infection on behavioural organization, the association between parasitism and complexity of activity patterns was compared between naturally infected sheep and experimentally deparasitized sheep. Possible changes in more traditional measures of behaviour were also tested as a function of sheep infection status. Finally, in an extra study, the potential impact of strongylid infection on the presence of malaria parasite’s DNA in ape faeces was explored, but no clear association between the two infections could be found. In both wild and domestic animal models, my overall results indicated significant variation in the temporal organization of behaviour in relation to strongylid infections. More complex feeding sequences were exhibited by chimpanzees with higher intensities of strongylid infections and strongylid-free sheep displayed a higher complexity in their activity sequences in comparison to infected sheep. Interestingly, no effect of parasite infection could be revealed by using more conventional analyses of behaviour. These findings support growing evidence that fractal analysis can be used as a sensitive method to monitor animal welfare status and could encourage future applications of complexity analyses to diagnose and monitor the trajectory of parasite infections.
