The effect of heavy metal ions on the development of Abies alba Mill. and Picea abies (L.) H. Karst. somatic embryos

Abstract:

Cílem této práce bylo sestavit testovací systém pro ohodnocení schopnosti Abies alba a Picea abies projít procesem somatické embryogeneze pod vlivem stresu z působení těžkých kovů. Embryogenní kultury různých buněčných linií obou druhů byly pěstovány na médiu obohaceném třemi různými koncentracemi (50, 250 a 500 µM) kadmia (Cd2+) a olova (Pb2+). Odezva byla zaznamenána ve stádiích proliferace, maturace a germinace. Dalším záměrem práce bylo posoudit vliv Cd2+ a Pb2+ na změny buněčných hladin adenozintrifosfátu (ATP) a dále detoxifikační potenciál embryogenních kultur A. alba a P. abies během 21-denní fáze proliferace.Testované těžké kovy významně omezily rychlost proliferace embryogenních kultur A. alba a P. abies. Nejvyšší testovaná koncentrace Cd2+ znatelně zpomalila nebo zastavila růst embryogenních kultur obou druhů. Proliferační poměr zůstal překvapivě vysoký v případě buněčných kultur P. abies ošetřených Pb2+ ve všech testovaných koncentracích. Během fáze maturace klesl celkový počet somatických embryí pod vlivem Cd2+. Schopnost tvorby raných pre-kotyledonárních a kotyledonárních somatických embryí u obou druhů byla podobně snížena, přičemž Cd2+ vyvolalo vyšší úmrtnost a bylo více toxické než Pb2+. Navíc se zvyšující se koncentrací těžkých kovů se úměrně zvýšila také hladina ATP. Embryogenní kultury A. alba a P. abies reagovaly na ošetření Cd2+ a Pb2+ stimulací tvorby fytochelatinů a jejich biosyntetických intermediátů. Se zvyšující se koncentrací těžkých kovů byl pro syntézu fytochelatinů využíván glutathion, což umožnilo vazbu k iontům těžkých kovů a tím zamezilo tvorbě reaktivních kyslíkových radikálů. Pb2+ u A. alba a Cd2+ u obou druhů vyvolaly nárůst všech antioxidačních thiolových sloučenin. U P. abies vyvolala nejnižší testovaná koncentrace Pb2+ nejvyšší antioxidační odezvu. Jelikož byl pozorován jen velmi nízký příjem Pb2+ do pletiv, lze vyšší odolnost P. abies přisuzovat jeho schopnosti omezit příjem Pb2+ po delší době působení. Výsledky působení Cd2+ u obou druhů a Pb2+ u A. alba nastiňují možnost testování využitelnosti těchto jehličnanů při fytoremediaci.

Abstract:

The aim of this work was to set up a test system for investigation of the ability of Abies alba and Picea abies to undergo somatic embryogenesis under the heavy metal stress. Early somatic embryos (ESEs) of different cell lines of both species were grown on media enriched with three different concentrations (50, 250, and 500 µM) of cadmium (Cd2+) and lead (Pb2+). Thus, the response was observed during proliferation, maturation and germination stages. Another aim of this work was to investigate the effects of Cd2+ and Pb2+ on changes of cellular levels of adenosine triphosphate (ATP) and detoxification potential of A. alba and P. abies ESEs throughout the 21-day proliferation period. The tested heavy metals significantly reduced the proliferation rate of A. alba and P. abies ESEs. The highest tested Cd2+ concentration markedly slowed or stopped the growth of ESEs in both species. Unexpectedly, the proliferation ratio remained fairly high for the P. abies cell lines treated with Pb2+ at all concentrations tested. During the maturation stage, the total number of somatic embryos declined under Cd2+ exposure. The formation of early-precotyledonary and cotyledonary somatic embryos in both species was similarly reduced, although Cd2+ caused a higher death rate and was more toxic than Pb2+. Moreover, by increasing concentration of the heavy metals, ATP levels were also proportionally increased. ESEs of A. alba and P. abies responded to treatment with Cd2+ and Pb2+ by inducing phytochelatins and their biosynthetic intermediates. By increasing heavy metal concentrations, glutathione was used for the synthesis of phytochelatins enabling the tissues to bind to heavy metal ions and thereby avoiding the production of reactive oxygen species (ROS). Pb2+ in A. alba and Cd2+ in both species caused similar increases of all antioxidative thiol compounds; thus, similar mechanisms involving a heavy metal-induced stress response can be assumed. In P. abies, the lowest Pb2+ concentration tested provoked the highest antioxidative response. Since a very low uptake of Pb2+ into the tissue was observed, the higher resistance of P. abies can be attributed to its ability to reduce Pb2+ uptake after longer exposure times. The results of Cd2+ treatment of both species and Pb2+ treatment of A. alba indicated the possibility of testing these coniferous species as potential phytoremediators.
