Influence of steroid hormones with a significant anabolic effect on the microscopic structure of pig muscle

Abstract:

Legislativa Evropské unie striktně zakazuje používání látek mající hormonální nebo thyreostatický účinek jako stimulátory růstu v krmivech pro hospodářská zvířata. Import masa ze zemí mimo EU je povolen pouze pod podmínkou doložení certifikace, že zvířatům nebyly anabolické látky podány a možný obsah reziduí těchto látek je monitorován. Problémem posledních let je aplikace „koktejlů“ širokého spektra anabolických látek o velice nízkých koncentracích, které jsou v potravinách těžce detekovatelné a je obtížné je kvantifikovat pomocí konvenčních detekčních metod. Je proto snaha o vývoj nových citlivých metod screeningu anabolik, nezávisle na jejich struktuře nebo koncentraci, založené na sledování biologických změn na úrovni genové exprese, změnách v metabolismu nebo struktuře na buněčné úrovni. V této práci byly sledovány histologické změny v mikrostruktuře svaloviny prasat, za účelem vytvoření nové cílené metodiky pro detekci anabolických steroidů v mase. Bylo využito 18 prasat rozdělených do tři skupin na základě rozdílné aplikace a dávkování steroidních léčiv: s aplikací 5 x 4 mg/kg testosteronu (T), s aplikací 5 x 5 mg/kg nandrolonu (N) a skupinu kontrolní (K). Po porážce byly ze svalu Musculus longissimus dorsi připraveny histologické preparáty, u kterých proběhlo hodnocení vybraných parametrů a obrazová analýza. Průměr svalových vláken byl zvýšen o 32 % ve skupině T, o 49 % ve skupině N, plocha průřezu svalovými vlákny byla zvýšena u T o 112 %, u N o 145 % ve srovnání s kontrolou. Množství endomysia mezi svalovými vlákny bylo ve skupinách T a N výrazně vyšší oproti kontrole. Množství jader ve svalových buňkách a množství kolagenních vláken se mezi skupinami statisticky nelišilo. Podávání anabolických steroidů tedy vyvolává výrazné změny v mikrostruktuře svaloviny prasat. V budoucnu by histologická analýza mohla být účinným dílčím nástrojem pro detekci anabolických látek nelegálně používaných při výkrmu.

Abstract:

European Union legislation strictly prohibits the use of preparations having a hormonal or thyrostatic effect as growth promoters in livestock feed. Imports of meat from countries outside the EU are allowed only under the condition of certification, that the animals have not been given anabolic substances. The possible content of residues of these substances is monitored. The problem of last years is the application of "cocktails" of anabolic substances with very low concentrations, which are difficult to detect in food and are difficult to quantify using conventional detection methods. Therefore, there is an effort to develop new sensitive methods of screening anabolics, regardless of their structure or concentration, based on monitoring biological changes at the level of gene expression, changes in metabolism or structure at the cellular level. In this work, histological changes in the microstructure of pig muscle were monitored in order to create a new targeted methodology for the detection of anabolic steroids in meat. The study included 18 pigs, divided into three groups based on different application and dosage of steroid drugs: with the application of 5 x 4 mg/kg testosterone (T), with the application of 5 x 5 mg/kg nandrolone (N) and the control group (K). After slaughter, histological specimens were prepared from the Musculus longissimus dorsi, in which the evaluation of selected parameters and image analysis took place. The diameter of muscle fibers was increased for 32% in group T, for 49% in group N, the cross-sectional area of muscle fibers was increased in T for 112%, in N for 145% compared to the control. The amount of endomysium between muscle fibers was significantly higher in groups T and N compared to the control. The number of nuclei in muscle cells and the amount of collagen fibers did not differ statistically between the groups. Thus, the administration of anabolic steroids causes significant changes in the microstructure of pig muscle. In the future, histological analysis could be an effective sub-tool for the detection of anabolic substances illegally used in fattening.
