Expression, purification, and characterization of intracellular regulatory domains of cytokinin receptor AHK4 in Arabidopsis thaliana.

Abstract:

The cytokinins are plant hormones, which regulate variety of biological functions. Their signaling is mediated via so called Multi-step phosphorelay, which consists of hybrid histidine kinase, histidine phosphotransfer protein and respond regulator. The cytokinins are perceived with three receptors in Arabidopsis thaliana: AHK2, AHK3 and AHK4. All three receptors are hybrid histidine kinases, which consist of sensor cytokinin-binding CHASE domain, histidine kinase domain with conserved His residue at which come to autophosphorylation, receiver-like domain, which biological function is unknown and receiver domain with conserved Asp residue, at which the phosphate group is transported from His and then transmitted to histidine phosphotransfer proteins. They act as transport mediators and transmit the phosphate into the nucleus, where the response regulators are located. They mediate the plant response to cytokinins. In this thesis, I tried to express and purify three fusion proteins containing the intracellular receiver-like domain of the receptor AHK4. The aim was to prepare the stabile and homogenous protein for further functional analysis of this domain, which function within the plants is still unknown. Unfortunately, I was unable to achieve the aim, because of the unsuccessfulness of obtaining native protein with the purification and cleveage of the fusion protein. In the thesis I also tested the function of C-terminal receiver domain of the receptor AHK4 with the binding study with the interaction partners from A. thaliana using ELISA. I tested the interaction of AHK4 with two histidine phosphotransfer proteins AHP2 and AHP3. AHK4 interacts in vitro with AHP3 and hardly interacts with AHP2. This findings correspond with the results obtained in yeast-two-hybrid system.

Abstract:

Cytokininy jsou rostlinné hormony zastávající různé biologické funkce. Jejich signalizace je sprostředkována tzv. vícekrokovou fosforylací, která se skládá z hybridní histidin kinázy, histidin fosfo-transferového proteinu a regulátora odpovědi. U Arabidopsis thaliana jsou cytokiníny vnímány třemi receptory: AHK2, AHK3 a AHK4. Všechny tři jsou hybridní histidin kinázy, které se skádají ze čtyř domén: senzorové CHASE domény vážící cytokininy, histidin kinázové domény, na které dochází k autofosforylaci konzervovaného zbytku His, pseudo-přijímačové domény s doposud neobjasněnou funkcí a přijímačové domény s konzervovaným zbytkem Asp, na který je přenášen fosfát z His a dále transmitován na histidin fosfo-transferové proteiny. Ty působí jako mediátory přenášející fosfát do jádra na regulátory odpovědi, které sprostředkůvají odpověď rostliny na cytokininy. Ve své diplomové práci jsem se pokusila exprimovat a purifikovat tři fúzní proteiny obsahující intracelulární pseudo-přijímačovou doménu receptoru AHK4. Cílem bylo připravit stabilní a homogenní protein pro funkční analýzu této domény, jejíž funkce u rostlin není doposud objasněna. Bohužel se mi nepovedlo tento cíl naplnit, jelikož ani purifikace ani štěpení fúzního proteinu pro získání nativního proteinu nebyly dostatečně úspěšné. V práci jsem také testovala funkčnost C-koncové přijímačové domény receptoru AHK4 pomocí vazebné studie s interakčními partneri z A. thaliana metodou ELISA. Testovala jsem interakci se dvěma histidin fosfo-transférovými proteiny AHP2 a AHP3. AHK4 interaguje in vitro s AHP3 a téměř neinteraguje s AHP2. Tyto výsledky jsou v souladu s výsledky získanými v kvasinkovém dvou-hybridním systému.
