Synthesis and biological study of new abscisic acid derivatives

Abstract:

Rostlinné hormony jsou látky regulující růst a vývoj rostlin. Mohou se vyskytovat ve volné formě nebo konjugované s různými substráty jako jsou aminokyseliny, cukry nebo peptidy. Konjugované rostlinné hormony mohou sloužit jako inaktivovaná, zásobní nebo transportní forma či dokonce bioaktivní látka. Rostlinný hormon kyselina abscisová (ABA) se podílí na regulaci mnoha fyziologických procesů, jako je např. dormance, klíčení semen, zrání nebo odpovědi na environmentální stresy. Dosud byl identifikován pouze jeden konjugát ABA, a to ABA-[beta]-D-glukopyranosyl ester (ABA-Glc). Todoroki a kol. (2011) provedli studii zaměřující se na syntetické konjugáty ABA s přirozenými aminokyselinami, ale žádná studie zabývající se konjugáty ABA s nepřirozenými aminokyselinami dosud nebyla publikována. Abychom prozkoumali možnou bioaktivitu takových látek, byla připravena malá série ABA konjugátů a jejich bioaktivita byla vyhodnocena v několika biotestech. Pouze derivát 6c vykazoval výraznou bioaktivitu podobnou efektu ABA. Bylo ale zjištěno, že tento derivát v rostlinách podléhá hydrolýze a jeho aktivita proto může být přisouzena ABA uvolněné hydrolýzou. Ostatní konjugáty nevykazovaly výrazný ABA agonistický efekt, ale u dvou z nich byla zjištěna schopnost ovlivňovat hladinu auxinů v rostlinách.

Abstract:

Plant hormones are substances regulating plant growth and development. They can occur in free form or conjugated to diverse substrates such as amino acids, sugars or peptides. Conjugated plant hormones can serve as inactivated, storage or transport or even as a bioactive substance. The plant hormone abscisic acid (ABA) is involved in regulation of many physiological processes such as dormancy, seed germination, ripening or responses to environmental stresses. Only one ABA conjugate, ABA-[beta]-D-glucopyranosyl ester (ABA-Glc) has been identified up to date. Todoroki et al. (2011) reported a study focusing on synthetic conjugates of ABA and natural amino acids, however, no study targeting conjugates of ABA and unnatural amino acid has been published. To examine possible action of such conjugates a small series of ABA conjugates was prepared within this thesis and their bioactivity in several bioassay was evaluated. Only derivative 6c displayed significant ABA-like effect. However, it was found out that it was subjected to hydrolysis within plants and therefore this derivative action can be attributed to ABA released by hydrolysis. The rest of the conjugates did not elicit significant ABA-like effect but two of them were shown to affect auxin levels in plants.
