Inductively coupled plasma mass spectrometry in the analysis of natural materials with a focus on geological and archaelogical samples

Abstract:

Předložená disertační práce je zaměřena na studium biominerálů a přírodních minerálů pomocí laserové ablace ve spojení s hmotnostní spektrometrií indukčně vázaného plazmatu (LA-ICP-MS). Kombinace vzorkování pomocí laserového svazku a ICP-MS jako detekčního systému předurčuje tuto moderní analytickou techniku k analýze pevných materiálů s cílem stanovit nejen obsahy matričních, minoritních, ale především stopových prvků, společně se studiem jejich 2D distribuce a vzájemné asociace ve studovaném systému. První část práce je věnována studiu biominerálů, konkrétně recentních a fosilních kosterních pozůstatků (kosti, zuby) a močových konkrementů. Multielementární analýza kosterních tkání poskytla informace, které byly využity k vyhodnocení paleo-redoxních podmínek během fosilizace, stanovení míry diageneze fosfátové fáze a/nebo změny mikrostruktury kostní tkáně v důsledku mikrobiální aktivity. V této části práce je diskutována problematika mapování a zobrazení povrchu prostřednictvím liniového skenu a diskrétních bodů společně s vhodnou kalibrační strategií. Druhá část práce zahrnuje aplikaci LA-ICP-MS v analýze geologických vzorků, horninotvorných minerálů – slída, živec, křemen, turmalín, s cílem definovat obsahy a asociace vybraných prvků. Detailně je zde diskutován proces interakce laserového svazku se studovaným materiálem, optimalizace metody pro analýzu minerálů a volba vhodného kalibračního materiálu, jakožto nejnáročnější a nejkritičtější části LA-ICP-MS experimentu. Studován byl vliv rozdílné orientace osy krystalu vybraných minerálů (slída a turmalín) na interakci s laserovým svazkem. Disertační práce zahrnuje také problematiku stanovení stopového množství cínu v geologických materiálech metodou LA-ICP-MS.

Abstract:

This dissertation thesis is focused on the study of biominerals and minerals using laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS). The combination of a laser beam sampling with an ICP-MS detection system categorizes this modern analytical technique for analysis of solids in order to determine not only the contents of matrix, minor, and especially trace elements, but also to investigate their 2D distributions and elemental associations in studied systems. The first part of the research is devoted to the study of biominerals, specifically recent and fossil skeletal remains (bones, teeth), and urinary stones. The multielemental analysis of skeletal tissues has provided the information used for evaluation of the paleo-redox conditions during fossilization, and also results in the determination of a degree of phosphate phase diagenesis as well as in changes in bone tissue microstructure due to microbial activity. 2D mapping via line scanning and discrete spot analysis is also included in this part. The attention has been paid to development of the suitable quantification procedure. The second part includes the application of LA-ICP-MS in the analysis of geological materials (rock-forming minerals – mica, feldspar, quartz, tourmaline) in order to define the chemical composition in the terms of contents of elements and their associations. The fundamental process of material-laser beam interaction including the careful optimizing the experimental parameters of LA as well as ICP-MS for analysis of minerals is discussed in more details. Beside methodology, the selection of a suitable calibration material has been investigated and assessed as the most demanding and critical step of the LA-ICP-MS analysis. The influence of different orientation of the crystal axis of mica and tourmaline on the signal stability and intensity with subsequent quantification has been studied. This dissertation thesis also includes the possibilities and limitations of tin determination in geological samples at the trace level by the LA-ICP-MS technique.
