A role of platelet mediators in modulation of functional response in professional phagocytes

Abstract:

Serotonin a histamin jsou důležité biogenní aminy uvolňované z aktivovaných krevních destiček. V místech zánětu se dostávají do bezprostředního kontaktu s ostatními buňkami imunitního systému. Porozumění interakcím mezi krevními destičkami a buňkami imunitního systému by mohlo vést k možnosti farmakologicky regulovat aktivitu profesionálních fagocytů spojenou s tvorbou kyslíkových a dusíkových metabolitů, a tím modulovat jejich patologické projevy. Předkládaná práce prokazuje inhibiční účinek serotoninu na produkci oxidu dusnatého myšími makrofágy i na proteinovou expresi inducibilní syntasy oxidu dusnatého. Serotonin byl také schopen inhibovat produkci prozánětlivých cytokinů, např. IL-6, TNF-α nebo IL-1. Účinek serotoninu na oxidativní vzplanutí makrofágů nebyl jednoznačně prokázán. Na druhou stranu u serotoninu byla prokázána výborná antioxidační kapacita. Dále byl pomocí kombinace histaminových agonistů a antagonistů prokázán inhibiční vliv histaminu na oxidativní vzplanutí fagocytů v plné krvi i samotných izolovaných lidských neutrofilů prostřednictvím histaminového H2R receptoru. Tato práce přispěla k objasnění mechanismů vlivu mediátorů krevních destiček na profesionální fagocyty a přinesla nové poznatky o úloze krevních destiček v zánětlivých stavech.

Abstract:

Serotonin and histamine are important biogenic amines released from activated platelets. These biogenic amines come into direct contact with the other immune cells at sites of inflammation. Understanding interactions between platelets and immune cells could lead to the possibility to pharmacologically regulate the activity of the professional phagocytes associated with the formation of oxygen and nitrogen metabolites and thereby to modulate their pathological manifestations. This work demonstrated the inhibitory effect of serotonin on nitric oxide production by murine macrophages and inducible nitric oxide synthase protein expression. Serotonin was also able to inhibit the production of pro-inflammatory cytokines such as IL-6, TNF-α or IL-1. The effect of serotonin on oxidative burst of macrophages has not been clearly demonstrated. On the other hand, serotonin has been demonstrated as an excellent antioxidant compound. Using a combination of histamine agonists and antagonists, inhibitory effect of histamine on the oxidative burst of phagocytic cells in whole blood and isolated human neutrophils via histamine H2-receptor was demonstrated. This thesis contributed to a clarification of mechanisms by which platelet-derived mediators affect the professional phagocytes and provides a new knowledge on the role of platelets and their mediators in inflammatory conditions.
