Iron chelators in oncology

Abstract:

Tato bakalářská práce se věnuje molekulárním mechanismům působení chelátorů železa a jejich využití v léčbě nádorových onemocnění. První část práce shrnuje metabolismus železa, jeho regulaci a změny tohoto metabolismu v nádorových buňkách. Druhá část představuje chelátory, tedy látky schopné vázat železo. Tato část se věnuje také potenciálnímu využití chelátorů k omezení přísunu železa k nádorovým buňkám, což může negativně ovlivňovat jejich metastatickou aktivitu a schopnost zvýšené proliferace. Poslední část se zaměřuje na schopnost chelátorů ovlivnit expresi řady genů, které kódují proteiny zapojené do rozličných signálních drah, zásadních pro rozvoj nádorového onemocnění. Přínos chelátorů v protinádorové terapii tak může být nejen na úrovni regulace množství železa, ale také díky působení na klíčově signální dráhy společně s dalšími chemoterapeutiky.

Abstract:

This bachelor thesis will discuss molecular mechanisms in the effects of iron chelators and their possible use in cancer treatment. The first part provides a summary of iron metabolism, its regulation and the changes of this metabolism in tumor cells. In the second part are presented chelators, which are agents able to bind iron. This part also describes their potential use in preventing iron from reaching tumor cells, which may negatively affect their metastatic activity and their ability of high proliferation. The last part focuses on the chelators’ ability to affect the expression of several genes which code proteins linked into various signal paths vital for the development of cancer disease. The contribution of chelators in anticancer therapy is possible not only through regulation of the amount of iron, but also through the effects on key signal pathways, together with other chemotherapeutics.
