A validation of the analytical procedure of the determination of AOX in the aqueous matrixes

Abstract:

V laboratorní praxi byla ověřena funkčnost komerčního kyvetového testu firmy Merck a skutečnost, zda tato metoda, kde místo klasického stanovení adsorbovatelných organicky vázaných halogenů metodou mikrocoulometrické titrace, jsou AOX stanoveny spektrofotometricky ve viditelné oblasti spektra, je vhodná k zavedení do běžné laboratoře. Metoda založená na spektrofotometrickém stanovení byla porovnávána s metodou mikrocoulometrické titrace. Z porovnání obou metod vyšla vítězně již zavedená a ověřená metoda mikrocoulometrické titrace, která je na rozdíl od spektrofotometrického stanovení velmi přesná. Jedinou její nevýhodou je vysoká pořizovací cena přístroje a nepřesnost při měření vysokých koncentrací. Naopak kyvetový test firmy Merck je ekonomicky výhodnější, nicméně se hodí pouze k orientačnímu zjišťování AOX ve vodách a je vhodný při stanovování vysokých koncentrací ve vodných matricích. Pro potřeby laboratoře byl vypracován standardní operační postup. Diplomovou prací byl zvalidován postup spektrofotometrického stanovení AOX testu firmy Merck.

Abstract:

Functionality of the commercial cell set of Merck company in laboratory practice and whether this method where adsorbable organically bound halogens are determined using spectro-photometric method in the visible spectrum zone is used instead of usually used micro-coulometric titration is suitable for introducion into a standard laboratory was verified. The method based on the spectro-photometric determination was compared with the method of micro-coulometric titration. The duel of both the methods has been won out through by the already introduced and proved method of micro-coulometric titration which is very accurate unlike the spectro-photometric determination. High purchase price of the instrument and inaccuracy when measuring in the field of high concentrations is the only disadvantage of this method. On the contrary, the cell test of Merck company is more advantageous when considering economical point of view. However, it is suitable only for approximate determination of AOX in water and when determining high concentrations in aqueous matrixes. A standard operation procedure has also been prepared. The dissertation has validated the procedure of spectro-photometric determination of AOX using the set of Merck company.
