Role of putative G-quadruplex forming sequeces in progression of neurodegenerative disorders

Abstract:

Hlavním cílem mé práce byl popis možného vlivu tvorby G-kvadruplexů na rozvoj neurodegenerativních chorob. Zaměřila jsem se na výskyt G4 sekvencí v genech u jednotlivých neurodegenerativních onemocnění, jejich schopnost tvořit G-kvadruplexy a popsat konkrétní role v těchto genech při rozvoji daného onemocnění. Závěr rešeršní části obsahuje doposud zjištěné možnosti terapeutického využití G-kvadruplexu u neurodegenerativních chorob. Experimentální část práce ověřuje tvorbu kvadruplexů v některých genech spojených s neurodegenerativními onemocněními pomocí metody EMSA.

Abstract:

The main goal of my thesis was the description of the possible influence of the formation of G-quadruplexes on the development of neurodegenerative diseases. I focused on the occurrence of G4 sequences in genes in individual neurodegenerative diseases, their ability to form G-quadruplexes, and the description of specific roles in these genes in the development of the given disease. The conclusion of the research part contains the possibilities of the therapeutic use of G-quadruplex in neurodegenerative diseases found so far. The experimental part of the work verifies the formation of quadruplexes in some genes associated with neurodegenerative diseases using the EMSA method.
