The MYB family proteins – their new functions in tumorigenesis and progression of malignant disease

Abstract:

Karcinom prsu představuje jeden z nejčastějších typů nádorových onemocnění diagnostikovaných u žen. Navzdory skvělým pokrokům, kterých bylo dosáhnuto v diagnostice a léčbě primárních nádorů, léčba metastatické formy tohoto onemocnění často selhává. Proto je potřeba zlepšit nejenom úspešnost léčby, ale nalézt taky spolehlivé prediktory metastatického šíření, které by byli použitelné v klinické praxi. Úloha transkripčního faktoru c-Myb je u prsního karcinomu považována za dvojakou, v závislosti na stádiu progrese nebo specifických aspektech mikroprostředí, c-Myb může působit jako faktor, který nádor podporuje nebo inhibuje. Exprese c-Myb je nejčastěji nalezena u prsních nádorů pozitivních na estrogenový receptor (ER), nicméně byla popsána taky napříč ostatními podtypy prsního karcinomu. Pro studium role proteinu c-Myb v prsním karcinomu jsme využili jeho nízké exprese u několika ER-negativních buněčných linií a připravili jsme jejich varianty s ektopickou expresí c-Myb. Naše stěžejní experimenty na myších modelech ukázali, že zvýšená exprese proteinu c-Myb snižuje kapacitu buněk prsního nádoru infiltrovat plíce. Proto jsme se zaměřili na mechanismus, kterým se protein c-Myb podílí na řízení metastatické aktivity buněk karcinomu prsu, zejména s ohledem na metastázy v plicní tkáni. Naším cílem byla identifikace té části transkripčního programu řízená proteinem c-Myb, která by mohla determinovat kapacitu buněk prsního karcinomu infiltrovat plíce. Porovnáním transkriptomů buněk 4T1 nadměrně exprimujících c-Myb a varianty buněk 4T1 vysoce účinně kolonizujících plíce jsme identifikovali sadu devíti genů (Ccl2, Cxcl1, Cxcl5, Cxcl16, Il1a, Lcn2, Icam1, Tnfrsf9, Ikbke), jejichž exprese je inhibována při zvýšené hladině proteinu c-Myb. Ukázali jsme, že exprese chemokinu CCL2 byla rozhodující pro úspěšnou transmigraci buněk prsního karcinomu přes plicní endotel pomocí monocytů. Popsali jsme cytokin IL1α jako klíčový aktivátor dráhy NF-κB v buňkách karcinomu prsu a ukázali jsme, že jeho transkripční inhibice řízená proteinem c-Myb může vést k potlačení exprese většiny genů z identifikované sady. Analýza klinických dat ukázala, že c-Myb a geny jejíchž exprese je ním potlačena jsou silným prediktorem relapsu plicních metastáz. Ukázali jsme, že nádory mléčné žlázy exprimující c-Myb jsou spojeny s nízkou infiltrací makrofágy asociovanými s nádory. Výsledky vedly ke vzniku tří původních vědeckých prací publikovaných v časopisech s impakt faktorem. Práce je prezentována jako komentovaný soubor těchto publikací.

Abstract:

Breast cancer (BC) represents one of the most common malignancies diagnosed in women. Despite the great advances achieved in the diagnosis and the cure of the primary BC lesions, the therapy of the metastatic disease often fails. Therefore, there is a necessity not only to improve the success rate of therapy but also to find reliable predictors of the metastatic spread usable in clinical practice. The role of the transcription factor c-Myb in BC is supposed to be dichotomous, according to the progression stage or specific microenvironmental aspects, c-Myb may act either as a tumor-promoting or tumor-suppressing factor. The expression of c-Myb is mostly found in the estrogen receptor (ER) positive BC, however, it is expressed across other BC subtypes as well. To study the c-Myb role in BC, we took advantage of its low expression in several ER-negative BC cell lines and prepared variants of these cells with c-Myb ectopic expression. Our pivotal experiments have shown that c-Myb overexpression diminished the lung infiltration capacity of BC cells in murine models. Therefore, we focused on the mechanism by which the c-Myb protein is involved in the control of metastatic activity of BC cells, especially concerning metastases in the lung tissue. We aimed to identify the part of the transcriptional program governed by c-Myb that could determine the lung infiltration capacity of BC cells. Comparing the transcriptomes of 4T1 cells overexpressing c-Myb and a variant of 4T1 cells highly proficient in the lung infiltration, we have identified a set of nine genes (Ccl2, Cxcl1, Cxcl5, Cxcl16, Il1a, Lcn2, Icam1, Tnfrsf9, Ikbke) whose expression is diminished upon the increased level of the c-Myb protein. We have shown that the expression of the CCL2 chemokine was decisive for successful monocyte-assisted transmigration of BC cells through the lung endothelium. We described the IL1α cytokine as the key NF-κB pathway activator in BC cells and showed that its transcriptional inhibition governed by the c-Myb protein may lead to suppression of most of the genes from the identified set. Analysis of clinical data has shown that c-Myb and its repressed genes are a strong predictor of relapse in lung metastasis. We have shown c-Myb expressing mammary tumors are associated with low tumor-associated macrophages infiltration. The results gave rise to three original scientific papers, published in journals with impact factor. The thesis is presented as a commented collection of these publications.
