Study of saccharide-recognising proteins and their role in pathogenesis

Abstract:

Lektiny tvoří významnou skupinu proteinů a glykoproteinů vyskytujících se nejen ve virech a bakteriích, ale také v rostlinách, houbách, plísních či živočiších. Díky své schopnosti specificky rozpoznávat a reverzibilně vázat glykokonjugáty se účastní celé řady fyziologických a patofyziologických procesů. Lektiny se mohou také podílet na interakcích patogenů s hostitelem, což může výrazně přispívat k rozvoji infekce. Stávají se tak dobrým cílem v antiadhezivní terapii, která by mohla vyřešit problém dnešní doby – rezistenci vůči antibiotikům. V současné době jsou lektiny nepostradatelným nástrojem v biotechnologii a lékařském výzkumu. Disertační práce je zaměřena na potenciální lektiny z bakterie Photorhabdus asymbiotica, konkrétně na nově identifikovaný lektin PHL. P. asymbiotica je oproti zbylým druhům rodu Photorhabdus nejen patogenem hmyzu, ale také patogenem člověka způsobujícím infekci měkkých tkání. Cílem této práce bylo detailně charakterizovat lektin PHL a následně otestovat jeho potenciální inhibitory. Charakterizace lektinu spočívala nejen ve stanovení jeho vazebných vlastností a struktury, ale také jeho vlivu na imunitní systém hostitele (jak hmyzího, tak lidského). K tomu byla využita celá řada metod, jako je např. afinitní chromatografie, rezonance povrchového plazmonu, izotermální titrační kalorimetrie, analytická ultracentrifugace, rentgenová krystalografie, hemaglutinace, fluorescenční mikroskopie, měření antimikrobiální aktivity, oxidativního vzplanutí a fenoloxidasové aktivity. Některé z těchto metod byly také využity pro stanovení inhibičního účinku syntetických fukosylovaných sloučenin založených na různých chemických strukturách. Práce je rozdělena na tři části. První je zaměřena na lektiny, sacharidy a rod Photorhabdus. V druhé části jsou popsány cíle projektu společně s dosaženými výsledky. Podstatná část naměřených dat již byla publikována, proto jsou shrnuty jako abstrakty k příslušným publikacím. Poslední část stručně shrnuje vědecké projekty, kterým byla také v průběhu doktorského studia věnováno značné úsilí.

Abstract:

Lectins are ubiquitous carbohydrate-binding proteins found in bacteria, plants, animals, fungi and viruses, where they play key role in various processes. Different affinity towards different ligands enables lectins to “read” the information stored in carbohydrate molecules, thus making them a powerful tool in a whole range of physiological or pathophysiological functions. Besides these functions, they also play a very important role in interactions between a pathogen and its host and thus can mediate the first step of an expansion of infection. It makes from them a potential target for glycomimetics and anti-adhesion therapy, overcoming antibiotic resistance related issues. Currently, they are essential tools in biotechnology and medical research. The dissertation was focused on studying one of the newly identified lectins from the Photorabdus asymbiotica bacterium – the PHL lectin. Unlike the other three species of the genus Photorhabdus, P. asymbiotica is not only an insect pathogen but can also infect humans and cause both locally invasive soft tissue and disseminated bacteraemic infection. The goal of this thesis was the first of all to completely characterize the PHL lectin and then to test synthetic mono- and multivalent ligands as potential inhibitors of PHL. The characterization comprised of both determination of binding properties and the structure and also observation of its effect on host immune system (both insect and human). Various methods were employed, e.g. affinity chromatography, surface plasmon resonance, isothermal titration calorimetry, analytical ultracentrifugation, X-ray crystallography, haemagglutination, fluorescence microscopy, measurement of antimicrobial activity, oxidative burst and phenoloxidase activity. Some of these methods were also utilized for determination of inhibition potency of synthetic fucosylated compounds based on different scaffolds. The thesis is divided into three parts. The first part discusses the general background of lectins and carbohydrates and also explains our interest in Photorhabdus genus. The next part provides the goals of the PHL project and describes acquired data. Essential part of the results has been already published therefore they are summarized as abstracts to each publication. The last part briefly sums up other scientific projects which were also paid attention during the doctoral study.
