Evaluation of repellency effect of selected essential oils against sheep tick (Ixodes ricinus)

Abstract:

V rámci této práce byly zhodnoceny repelentní účinky třech esenciálních olejů proti nymfám klíštěte Ixodes ricinus. Silice z Rosmarinus officinalis, Salvia officinalis a Picea abies byly testovány v koncentracích 5 %, 10 %, 15 % a 20 %. Klíšťata potřebná pro pokus byla sbírána v Praze v Kunraticko-michelském lese v okolí Kunratického potoka a v Rakovníku od června do září roku 2019. Pro pokus byla celkem potřeba 2310 nymf, které byly po ukončení testu usmrceny mrazem při -30 °C. Samotný test repelentní účinnosti byl prováděn in-vitro v Petriho misce (Ø 90 mm) s třemi vrstvami filtračního papíru, kdy se na jeho okraj aplikovalo 0,5 ml etanolového roztoku esenciálních olejů o příslušné koncentraci. Po zaschnutí repelentu byla do experimentální arény vložena klíšťata vždy po 10 kusech a ve třech opakováních. Účinnost byla sledována v časech: 5 minut, 30 minut, 1 hodina, 2 hodiny, 4 hodiny, 6 hodin a 8 hodin od aplikace. Test byl ukončen také v případě, že repelence byla nižší než 33 %. Na závěr byly stejným postupem otestovány 5% syntetický repelent DEET a Alpa Francovka s hřebíčkem, tedy tzv. domácí repelent vyrobený dle návodu doporučovaného českými elektronickými i tištěnými médii. Vědecká hypotéza „Testované esenciální oleje mají určitý repelentní účinek, není ovšem tak stabilní jako u syntetických preparátů“ byla potvrzena, když 5% repelent DEET vykazoval po celou dobu testování signifikantně (α = 0,05) nejvyšší repelenci než testované esenciální oleje ve všech koncentracích. Na základě statistického hodnocení bylo také zjištěno, že na hladině α = 0,05 byla účinnost ovlivněna typem repelentu, a to po celou dobu testování. Od 1 hodiny po aplikaci pak byla účinnost signifikantně ovlivněna i koncentrací repelentu. Co se týče porovnání účinku esenciálních olejů, jako nejlepší se z testovaných jevil smrkový (Picea abies) olej v koncentraci 20 %.Na základě získaných výsledků lze konstatovat, že všechny testované silice měly určitý repelentní účinek, který lze zvýšit zvýšením koncentrace. Tento účinek je ovšem v porovnání se syntetickým repelentem DEET nepříliš stabilní. Na druhou stranu se prokázalo, že zejména šalvějový a smrkový esenciální olej je při opakované aplikaci možné považovat za alternativu pro běžné syntetické repelenty.

Abstract:

In this work, the repellency effects of three essential oils against the nymphs of the tick Ixodes ricinus were evaluated. Essential oils from Rosmarinus officinalis, Salvia officinalis and Picea abies were tested at the concentrations of 5%, 10%, 15% and 20%. The ticks for the experiment were collected in Prague in the Kunratice-Michle forest near the Kunratický stream and in Rakovník in the period of June to September 2019.Totally, 2310 nymphs were required for the experiment. The ticks were killed by freezing at -30 ° C at the end of the test. The repellency was tested in-vitro in using Petri dishes (Ø 90 mm) with three layers of filter paper. The periphery of such arenas were treated by 0.5 ml of an ethanolic solution of essential oils. After the repellent had dried, 10 ticks were inserted into the experimental arena. Each trial was performed in three replicates. The efficiency was monitored 5 minutes, 30 minutes, 1 hour, 2 hours, 4 hours, 6 hours and 8 hours after the application. The test was also terminated if the repellency was less than 33 %. Finally, the 5% synthetic repellent DEET and solution of Alpa Francovka with clove (i.e. the so-called home repellent produced according to the instructions recommended by the Czech electronic and printed media) were tested in the same way.The scientific hypothesis "Tested essential oils have a certain repellency effect, but not as stable as the synthetic products" was confirmed when 5% DEET repellent showed significantly (p < 0.05) highest repellency than tested essential oils at all the concentrations. Based on the statistical evaluation, it was also found that (at the level of α = 0.05) the efficiency throughout the testing was affected by the repellent type. Excluding intervals of 5 and 30 minutes after the application, the efficiency was also significantly affected by repellent concentration. Regarding tested essential oils, spruce (Picea abies) oil in a concentration of 20 % seemed to be the best alternative repellent against tick nymphs.Based on the obtained results, it can be concluded that all the tested essential oils had certain repellency effect, which can be improved by concentration increase. However, this effect is not very stable compared to the synthetic DEET repellent. On the other hand, it has been shown that particulary sage and spruce essential oils might be considered as an alternative to conventional synthetic repellents when applied repeatedly.
