Allenyl synthon in synthesis of adamantane substituted heterocycles

Abstract:

V této práci je popsána příprava různě substituovaných nesymetrických allenylazinů obsahujících ve své struktuře substituent adamantyl. Tyto látky byly následně využity v termicky iniciovaných intramolekulárních cyklizačních reakcích za vzniku nových substituovaných 5,5-dimethyl-5,6- dihydro-4H-pyrrolo[1,2-b]pyrazolů. Allenylaziny v přítomnosti dipolarofilu vstupovaly do kombinované intra-intermolekulární criss-cross cykloadice za vzniku nových kondenzovaných tricyklických heterocyklů s různou substitucí v polohách 6 a 9. Toto zadání vychází ze znalostí o účinku adamantanového skeletu na biologickou účinnost, zejména zlepšení průniku molekuly s tímto skeletem buněčnou stěnou.

Abstract:

The Theses describes preparation of variously substituted nonsymmetrical allenylazines containing adamantyl. These compounds underwent thermally initiated intramolecular cyclizations leading to substituted 5,5-dimethyl- 5,6-dihydro-4H-pyrrolo[1,2-b]pyrazoles. Allenylazines in the presence of dipolarophile entered combined intra-intermolecular criss-cross cycloaddition reactions affording new fused tricyclic heterocycles with various substitutions in positions 6 and 9. The project was based on the knowledge of adamantane skeleton biological activity, which causes better transfer of molecules with this skeleton through cell membranes.
