Diversity of trypanosomatids in insects from Central African Republic

Abstract:

Trypanosomatids are flagellated protists threatening humans, animals and plants almost all over the worlds. These parasites have large diversity and many species still remains undiscovered. Studying diversity helps to map these species as well as elucidate evolution of parasitism, hosts' specificity or endosymbiotic relationships. This work deals with diversity of insects' trypanosomatids. Direct PCR amplification of trypanosomatid ITS1 and 18S rRNA gene from infected host gut, molecular cloning (in case of mixed infection), and sequencing of samples were used. Phylogenetic analysis using obtained sequences revealed 11 known and 23 new typing units, most of which were distributed over the majority of trypanosomatid lineages including uncharacterized ones. Two typing units were proved to be non-trypanosomatid species from the related order Parabodonida.

Abstract:

Trypanosomatidae je skupina bičíkatých protistů ohrožujících lidi, zvířata a rostliny téměř po celém světě. Tito parazité mají obrovskou diverzitu a mnoho druhů pořád zůstává neodhaleno. Studium diversity napomáhá mapovat tyto druhy, stejně tak pomáhá vysvětlit evoluci parazitismu, hostitelskou specifitu nebo endosymbiotické vztahy. Tato práce se zabývá diverzitou trypanosomatid hmyzu. Byly použity molekulární metody jako PCR amplifikace ITS1 a 18S rRNA genu trypanosomatid z infikovaných střev hostitelů, klonování (v případě mixovaných infekcí) a následná sekvenace vzorků. Fylogenetická analýza odhalila 11 známých a 23 nových typových jednotek, z nichž většina byla distribuována ve většině linií trypanosomatid, včetně těch necharakterizovaných. Dvě typové jednotky nepocházely z trypanosomatid, ale od jejich příbuzného řádu Parabodonida.
