Origin and coexistence of the spined loach populations (Cobitis) in the Po Valley

Abstract:

Sekavci rodu Cobitis jsou při dně a v substrátu žijící sladkovodní ryby. Středoevropské druhy mezi sebou hybridizují za vzniku gynogeneticky se rozmnožujících hybridních linií, z nichž většina je polyploidních. Hybridní linie expandovaly napříč areály rodičovských druhů a dokázaly rovněž sexuálně parazitovat na nerodičovském druhu. Předkládaná práce studovala 7 populací sekavců v povodí italské řeky Pád, odkud je známý endemit C. bilineata. Vedle očekávaného morfotypu C. bilineata odhalily morfologické analýzy také druhý morfotyp podobný druhu C. elongatoides a jeho syntopických hybridů. Kombinace genu mitochondriální DNA pro cytochrom b a jaderného DNA markeru (první intron genu pro S7) spolu s morfologií potvrdila výskyt obou pohlaví C. bilineata na všech lokalitách. Genetická analýza současně rozšířila informaci o vnitrodruhové variabilitě. Druhým, dosud jižně pod Alpami a v úmoří Jaderského moře neidentifikovaným taxonem, byli na pěti lokalitách sekavci hybridního původu. Mitochondriální haplotyp poukazoval na tzv. Hybridní klád I, známý především z povodí Dunaje. Somatický genotyp hybridů obsahoval vždy minimálně dvě sady genomu C. elongatoides a jednu sadu genomu C. tanaitica. Spojení biologických dat s genetickými naznačuje, že by mohlo jít o polyploidní gynogeneticky se rozmnožující linie, které zřejmě pro své rozmnožování využívají místní druh C. bilineata. Výsledky biologické rozmanitosti sekavců v povodí řeky Pádu s potvrzením nových biotypů mne vedou k zájmu pokračovat ve studiu těchto sekavců ve snaze porozumět jak se hybridi do oblasti Pádu dostali, jakým způsobem se rozmnožují a nakolik jsou zde geograficky rozšířeni a početní.

Abstract:

Spined loaches of the genus Cobitis are freshwater fishes living by the river bottom and in the substrate. The Central European species hybridize and giving rise gynogenetic hybrid lineages, which most of them are of polyploid constitution. Hybrid lineages expanded across the parental taxa distribution area and they also sexually parasitize some nonparental taxa. This Thesis studied seven populations of spined loaches from the Po River Basin, from where the endemic C. bilineata is known. Morphological analyses detected the expected morphotype of C. bilineata but also the second morphotype resembling the species C. elongatoides and its syntopic hybrids. The combination of the mitochondrial cytochrome b gene and the nuclear DNA marker (the first intron of S7) with morfological analysis confirmed the presence of both sexes of C. bilineata at all localities. The genetic analysis provided also some new information about the intraspecific variation. The second morphotype represented spined loaches of the hybrid origin which were never identified southward of the Alps and in the Adriatic Sea Basin before. These hybrids were discovered at five out of seven localities. Hybrids share the mitochondrial haplotype belonging to the Hybrid clade I, that is known mainly from the Danube Basin. The somatic genotype of hybrids were composed at least of two sets of C. elongatoides genome and one set of C. tanaitica genome. A combination of biological and genetic data indicate these might be polyploid gynogenetic lineage which sexually parasitize a local C. bilineata. My goal is to understand how the hybrids have appeared in the region in question, how they reproduce, and what id theis overal ditribution and population size.
