GLUT-1 Deficiency Syndrome

Abstract:

Tato bakalářská práce podává ucelený přehled o GLUT1 deficiencí, vzácném genetickém onemocnění způsobeném mutací v genu SLC2A1, který narušuje klíčový transport glukózy přes hematoencefalickou bariéru, což vede k energetickému deficitu mozku a následným neurologickým potížím, jako jsou epilepsie či psychomotorická retardace. Práce se zaměřuje na patofyziologii, molekulárně-genetické aspekty ovlivňující variabilitu příznaků, diagnostiku a současné možnosti léčby s důrazem na ketogenní dietu, přičemž hlavním cílem je poukázat na zásadní význam včasné diagnózy a terapie pro zlepšení kvality života pacientů.

Abstract:

This bachelor's thesis provides a comprehensive overview of GLUT1 deficiency, which is a rare genetic disorder caused by a mutation in the SLC2A1 gene. This mutation impairs the crucial transport of glucose across the blood-brain barrier, leading to cerebral energy deficits and subsequent neurological impairments, such as epilepsy and psychomotor retardation. The thesis focuses on the disorder's pathophysiology and the molecular genetic aspects that influence clinical variability. It also covers diagnostics and current treatment options, particularly the ketogenic diet. The primary aim is to emphasise the importance of early diagnosis and appropriate therapy in significantly improving patients' quality of life.
