Analýza sérových mikroRNA u pacientů s kolorektálním karcinomem

Analysis of microRNAs in serum of colorectal cancer patients

Abstract:

Východisko: Kolorektální karcinom (CRC) patří mezi nejčastěji se vyskytující nádorová onemocnění v ČR i celé západní Evropě. Úmrtnost dosahuje u tohoto onemocnění téměř 50%, a to především díky pozdní diagnostice onemocnění. Z toho důvodu je snaha o zavedení nové neinvazivní metody, která by dokázala identifikovat rané stádium onemocnění, které je ještě dobře léčitelné. Metody: Pomocí metody qRT-PCR byly změřeny hladiny miR-16, miR-17-3p, miR 29a, miR-92a a miR 135b na 100 vzorcích séra pacientů ve všech 4 stádiích kolorektálního karcinomu a na 31 vzorcích zdravých jedinců. Jako referenční gen byla zvolena miR-16. Pomocí Mann-Whitneyho testu pak byla porovnána exprese jednotlivých miRNA u pacientů a zdravých jedinců. Dále byla srovnána hladina miR 17-3p, miR-29a, miR-92a, miR 135b v jednotlivých klinických stádiích onemocnění pomocí Kruskal-Wallisova testu. Výsledky: U žádné z měřených miRNA - miR 17-3p (P = 0,65), miR-29a (P = 0,27), miR-92a (P = 0,90), nebyla naměřena statisticky významná změna hladiny v séru pacientů s CRC. MiR-135b nebyla v séru detekována. Porovnání exprese v jednotlivých klinických stádiích CRC mělo rovněž negativní výsledky pro miR 17-3p (P = 0,72), miR-92a (P = 0,16) a miR 135b (P = 0,87). U miR-29a byla nalezena korelace zvyšující se hladiny s vyšším klinickým stádiem (P = 0,045). Závěr: V této práci nebyl potvrzen předpoklad, že hladina analyzovaných miRNA v séru u pacientů s CRC by měla být pozměněna oproti hladině v séru zdravých dárců. Korelace mezi vzrůstajícím klinickým stádiem CRC a zvyšující se hladinou miRNA v séru byla potvrzena u miR-29a, přestože hladina miR-29a v séru nebyla potvrzena jako signifikantně zvýšená. U ostatních miRNA nebyla korelace stádia CRC na hladině miRNA v séru nelezena. Dle těchto výsledků by nebylo vhodné používat ani jednu z testovaných miRNA při diagnostice CRC

Abstract:

Objective: Colorectal carcinoma (CRC) is one of the most frequent malignant diseases in the Czech Republic, and even in whole western Europe. Mortality of this type of disease is almost 50%, primarily due to late diagnosis. A result of this fact is an effort to introduce a new non-invasive method that would be able to identify early stage of disease which is still successfully curable. Methods: The qRT-PCR method has been used to measure the level of miR-16, miR 17-3p, miR 29a, miR-92a and miR 135b of 100 samples of serum of patients in all 4 stages of colorectal carcinoma, and of 31 samples of the healthy ones. MiR-16 has been chosen as a reference gene. Using Mann-Whitney test, the expression of individual miRNAs of patients and healthy persons has been compared. Then, using Kruskal-Wallis test, it has been also compared the level of miR 17 3p, miR-29a, miR 92a, miR 135b in individual clinical stages of disease. Results: None of measured miRNAs - miR 17-3p (P = 0,65), miR-29a (P = 0,27), miR-92a (P = 0,90) had statisticaly significant altered level in serum of patiens with colorectal carcinoma. MiR-135b has not been detected in serum. A result of comparison of expressions in individual clinical stages of colorectal carcinome has been also negative for miR 17 3p (P = 0,72), miR-92a (P = 0,16) and miR 135b (P = 0,87). However, miR 29a has showed a correlation of increasing level with higher clinical stage (P = 0,045). Conclusion: This work has not confirmed an assumption that the level of analyzed miRNA in serum of CRC patients should have been altered in contrast to the serum of healthy donors. Corelation between increasing clinical stage of CRC and increasing level of miRNA in serum has been validated for miR-29a despite the fact that miR-29a has not been significantly increased in serum. Corelation of CRC stage with higher level of miRNA in serum has not been found concerning the other miRNAs. According to these results it would not be convenient to use any of tested miRNA for CRC diagnosis.
