Germination of peas with abiotic stress.

Abstract:

V této bakalářské práci byl sledován vliv abiotických stresů sucha a zasolení na klíčení semen hrachu. Hrách setý (Pisum sativum) patří do čeledi bobovité (Fabaceae). Je to jednoletá luskovina, typická svojí tvrdosemenností a hypogeickým klíčením. V teoretické části byla pospána charakteristika rostlin, semen, klíčení, dormance a stresů. Praktická část byla zaměřena na vlastní výzkum, kde byl uveden postup, materiály a pomůcky. Dále byl vysvětlen výpočet sledovaných parametrů pro testy klíčivosti. Cílem práce bylo vyhodnotit vliv stresorů na klíčení a objasnit hypotézu pozitivního působení peroxidu vodíku a omeprazolu při těchto stresujících podmínkách. Základním předpokladem bylo snížení klíčivosti a omezení růstu kořínku i nadzemní části semen v nepříznivém prostředí. Experimentální část probíhala v laboratořích na České zemědělské univerzitě v Praze. Deficitu vody bylo dosaženo pomocí PEG 6000 (koncentrace 15; 30; 45 mM) a salinity díky NaCl (50; 100;150 mM). Klíčení probíhalo v Petriho miskách vyložených filtračním papírem při optimálních podmínkách (konstantní teplota 25 C a vlhkost 60 %). V první části experimentu bylo založeno 6 variant: 50 mM NaCl; 100 mM NaCl; 150 mM NaCl; 15 mM PEG; 30 mM PEG; 45 mM PEG. V druhé části experimentu se pracovalo se 7 variantami: H2O; H2O2; OMP; 150 mM NaCl + H2O2; 30 mM PEG + H2O2; 150 mM NaCl + OMP; 30 mM PEG + OMP. V pokusu byla sledována klíčivost, energie klíčení, rychlost klíčení a růstové parametry klíčících semen. Z výzkumu vyplývá, že vodní deficit i salinita negativně ovlivňovaly klíčení semen a inhibovaly růst sazenic. Se zvyšující koncentrací PEG i NaCl docházelo k snižování sledovaných parametrů semen. Semena hrachu se jevila citlivější k deficitu vody než k zasolení. Nejhorších výsledků dosáhla varianta 45 mM PEG, která byla svojí koncentrací pro klíčení inhibující. Ošetření H2O2 či OMP ovlivnilo při stresových podmínkách některé parametry pozitivně, jiné negativně. Stresy jsou v zemědělství celosvětovým problémem, a je proto nutné se jimi zabývat a zkoumat látky, které by jejich negativní vliv na rostlinu snížily, a tak pozitivně ovlivnily jejich vývoj.

Abstract:

In this bachelor's thesis, the effect of abiotic stresses of drought and salinization has been observed on the germination of pea seeds. Garden pea (Pisum sativum) belong to the legume family (Fabaceae). It is an annual legume, characterized by its hardness and hypogeal germination. The characteristics of plants, seeds, germination, dormancy and stress were described in the theoretical part. The practical part was focused on the research, where the procedure, materials and tools were presented. Furthermore, the calculation of the monitored parameters for germination tests was explained. The aim of the work was to evaluate the effect of stressors on germination and to clarify the hypothesis of a positive effect of hydrogen peroxide and omeprazole under these stressful conditions. The basic premise was to reduce germination and limit the growth of the root and the aboveground part of the seeds in an unfavourable environment.The experimental part took place in laboratories at the Czech University of Life Sciences in Prague. The deficit of water was achieved with PEG 6000 (concentration 15; 30; 45 mM) and the salinity with NaCl (50; 100; 150 mM). Germination took place in Petri´s dishes lined with filter paper under optimal conditions (constant temperature 25 °C and 60 % of humidity). In the first part of the experiment, 6 variants were established: 50 mM NaCl; 100 mM NaCl; 150 mM NaCl; 15 mM PEG; 30 mM PEG; 45 mM PEG. In the second part of the experiment, 7 variants were used: H2O; H2O2; OMP; 150 mM NaCl + H2O2; 30 mM PEG + H2O2; 150 mM NaCl + OMP; 30 mM PEG + OMP. Germination, germination energy, germination rate and growth parameters of germinating seeds were monitored in the experiment. The research shows that water deficit and salinity negatively affected seed germination and inhibited seedling growth. With increasing concentration of PEG and NaCl, the monitored seed parameters decreased. Pea seeds appeared to be more sensitive to water deficit than to salinity. The worst results were obtained with the 45 mM PEG variant, which was inhibitory in germination concentration. H2O2 or OMP treatment affected some parameters positively under stress conditions, others negatively. Stress is a global problem in agriculture and it is therefore necessary to address it and study substances that would reduce their negative impact on the plant and in this way positively affect their development.
