Staphylococcus aureus as a significant alimentary pathogen

Abstract:

Cílem práce je charakterizovat bakterii Staphylococcus aureus a následně podpořit teoretickou část práce vlastním mikrobiologickým stanovením bakterie ve vybraných vzorcích. Teoretická část práce je zaměřena na celkové shrnutí dostupných informací o této bakterii. V práci je popsán význam bakterie, faktory ovlivňující růst, virulentní faktory a patogenita, která souvisí s podrobněji charakterizovaným alimentárním onemocněním vyvolávaným právě stafylokokovými enterotoxiny. Jsou zde zmíněna také další stafylokoková onemocnění, například dermatitidy či syndrom toxického šoku, a je objasněna i problematika rezistence k antibiotikům. Důležitým tématem je výskyt S. aureus v potravinách zapříčiněný převážně kontaminací lidmi a také možnosti včasné detekce bakterie. Experimentální část práce je zaměřena na detekci a stanovení S. aureus ve vzorcích klobás a sýrů pomocí různých kultivačních metod. Výsledky analýzy klobás na Baird-Parker agaru byly zjištěny dle očekávání, některé vzorky byly pozitivní na S. aureus, neboť došlo ke kontaminaci při manipulaci s potravinou. Analýza sýrů na třech typech médií – BPA půdě, MicroFast® a Petrifilm™ – vyloučila jeden kontaminovaný vzorek neodpovídající kvality, další výsledky byly v toleranci. Při srovnání spolehlivosti kultivačních metod bylo jako nejspolehlivější jednoznačně označeno standardní stanovení na BPA půdě, následně Petrifilm™ a jako poslední MicroFast®.

Abstract:

The aim of the thesis is to characterize the bacterium Staphylococcus aureus and then to support the theoretical part of the thesis by the actual microbiological determination of the bacterium in selected samples. The theoretical part of the thesis is focused on a general summary of the available information about this bacterium. The significance of the bacterium, factors influencing growth, virulence factors and pathogenicity are described in the thesis. Pathogenicity is closely related to a more detailed characterization of the alimentary disease caused by staphylococcal enterotoxins. Other staphylococcal diseases, such as dermatitis and toxic shock syndrome, are also mentioned, and the issue of antibiotic resistance is clarified. An important topic is the occurrence of S. aureus in food, mainly due to human contamination, and the possibilities for early detection of the bacterium. The experimental part of the work is focused on the detection and determination of S. aureus in sausage and cheese samples using different culture methods. The results of the analysis of sausages on Baird-Parker agar were as expected, with some samples being positive for S. aureus due to contamination during food handling. Analysis of cheeses on three types of media – BPA medium, MicroFast® and Petrifilm™ – excluded one contaminated sample of inadequate quality, other results were within tolerance. When comparing the reliability of the culture methods, the standard assay on BPA medium was clearly identified as the most reliable, followed by Petrifilm™ and lastly MicroFast®.
