Abstract:

Rostliny jsou přisedlé organismy, a proto si pro zvládnutí nepříznivého životného prostředí vyvinuly široký spektrum mechanismů, jak zvládnout nepříznivé prostředí. Biotické a abiotické faktory ovlivňují růst a formují metabolismus rostlin tak, aby podporoval jejich přežití v neoptimálních podmínkách. Tato práce s názvem „Změny metabolomu rostlin v reakci na kombinaci biotického a abiotického stresu“ charakterizuje účinky endofytické houby Acremonium alternatum na modelovou rostlinu Arabidopsis thaliana při stresu ze zasolení. Teoretická část shrnuje účinky sucha, extrémních teplot a zasolení na metabolismus rostlin a nastiňuje interakci rostlin s endofytickými organismy. Výsledky jsou rozděleny do tří částí. Nejprve byly analyzovány účinky zasolení na růst a metabolom Arabidopsis. Dále byly vybrány koncentrace solí odpovídající mírnému a silnému stresu a rostliny byly inokulovány A. alternatum. Výsledky ukazují, že endofytická houba A. alternatum podporovala odolnost a zlepšovala růst rostliny. Nakonec byla analyzována předpokládaná role signalizace cytokininů ve sledované prospěšné interakci. Výsledky však nebyly přesvědčivé a k řešení této otázky bude zapotřebí více biologických opakování.

Abstract:

Plants are sessile organisms and have evolved a broad spectrum of mechanisms to cope with the unfavorable environment. Biotic and abiotic factors influence growth and shape plants' metabolism to promote their survival under suboptimal conditions. This thesis entitled "Plant metabolome changes in response to a combination of biotic and abiotic stress" characterizes the effects of endophytic fungi Acremonium alternatum on the model plant Arabidopsis thaliana under salinity stress. The theoretical part summarizes the effects of drought, heat, and salinity on plant metabolism and outlines plant interaction with endophytic organisms. Results are separated into three parts. First, the effects of salinity on Arabidopsis growth and metabolome were analyzed. Next, the salt concentrations corresponding to low-level and severe stress were selected, and plants were inoculated with A. alternatum. Results indicate that A. alternatum promoted the plant's resilience and improved its growth. Finally, the putative role of cytokinin signaling in the observed beneficial interaction was analyzed. However, the results were inconclusive, and more biological replicates will be required to address this question.
