The effect of N(6)-substituated adenosine derivatives on proliferation and cell death in tumour cells

Abstract:

Tato bakalářská práce pojednává ve své první části o buněčné smrti a procesu apoptosy a jejích hlavních mechanismech. Druhá část se věnuje problematice purinů a především adenosinu s důrazem na jeho signální funkci, extracelulární a intracelulární metabolismus a úlohu při indukci buněčné smrti. Následně se zaměřuje na problematiku adenosinových derivátů, včetně klinické aplikace a na specifitu N6-substituovaných derivátů adenosinu. Praktická část práce řeší otázku in vitro aplikace tří vybraných N6-substituovaných adenosinových derivátů: N6-benzyladenosinu, N6-(3-iodobenzyl)adenosinu a 2-chloro-N6-(3-iodobenzyl)adenosinu na lidských leukemických buněčných liniích HL-60, U-937, K-562, K-562/DOX. Zaměřuje se na změny cytotoxický vlastností v důsledku rozdílné struktury daných derivátů a indukci apoptosy, jejíž projevy sleduje metodami založenými na morfologických a biochemických změnách způsobených v důsledku tohoto procesu

Abstract:

This bachelor thesis describes in the first part the cell death and the general aspects and the signalling pathways of the apoptosis process. The second part attends to purines with the focus on their signal function and extracellular and intracellular metabolism as a part of the cell death. Consequently, the work gives an attention to the derivatives of adenosine and the specifity of the N6-substituated included their clinical applications. Experimental part investigates the in vitro use and application of the three N6-substituated derivatives of adenosine: N6-benzyladenosine, N6-(3-iodobenzyl)adenosine and 2-chloro-N6-(3-iodobenzyl)adenosine in human leukaemia tumour cells HL-60, U-937, K-562, K-562/DOX. Laboratory work focuses on the modification of cytotoxic properties as a result of the different molecular structure of the derivatives and investigation of apoptosis with the techniques based on the morphological and biochemical changes during the apoptotic process
