Studies of functions of thioredoxin reductase in development and antioxidant system of honey bee

Abstract:

Teoretická část diplomové práce na úvod shrnuje základní poznatky o včele medonosné, reaktivních formách kyslíku a antioxidační obraně hmyzu. Dále je zaměřena na enzymy thioredoxinreduktasy (TrxR) a jejich význam. Thioredoxinreduktasy tvoří společně s thioredoxiny thioredoxinový systém, který je v organismu zapojen do mnoha různých funkcí. Za nejvýznamnější funkci u hmyzu, jež postrádá enzym glutathionreduktasu (GR) se považuje antioxidační obrana, ovšem podílí se mimo jiné i na buněčné signalizaci a udržování redoxní rovnováhy. Hmyzí TrxR se odlišují od savčích především ve struktuře aktivního místa, a to přítomností páru Cys Cys namísto savčího páru Cys-Sec. Tato změna ovlivňuje reaktivitu a tím i funkce těchto příbuzných enzymů. V rámci praktické části byly testovány metody stanovení aktivity TrxR. Byla určena specifická aktivita a úplná inhibice methylviologenem u TrxR z Drosophila melanogaster (DmTrxR) a Rattus rattus (rTrxR) pomocí spektrofotometrické metody se substrátem DTNB. Aktivitu těchto enzymů ovšem nebylo možné stanovit fluorimetricky s využitím fluorescenčně značeného insulinu (FITC-insulinu) jako substrátu, a to nejspíše kvůli nevhodné přípravě FITC-insulinu. Dále byla optimalizována metoda detekce aktivity TrxR a GR pomocí specifického barvení gelů po nativní PAGE, přičemž tento způsob detekce se zdá být nevhodný pro GR. Nakonec byla sledována exprese TrxR z Apis mellifera pomocí metody qPCR. U včel krmených lipopolysacharidem o koncentraci 30 a 60 g?ml 1 byl zjištěn signifikantní pokles exprese TrxR, což mohlo být důsledkem řízeného zvýšení hladiny ROS ve střevech za účelem boje s patogeny. K pochopení vlivu stresových faktorů na funkci TrxR je ovšem potřeba provést další měření.

Abstract:

The theoretical part of the thesis summarizes the knowledge about honey bees, reactive oxygen species and antioxidant defence of insects. It also focuses on the enzymes thioredoxin reductase and their significance. Thioredoxin reductases along with thioredoxins form the thioredoxin system, which is involved in many different functions in the organism. In insects lacking the glutathione reductase enzyme, antioxidant defence is considered to be the most important function, but it is also involved in cell signalling and maintenance of redox balance, among other things. The main difference between insect and mammalian thioredoxin reductases is the structure of the active site, namely the presence of a Cys-Cys pair instead of the mammalian Cys-Sec pair. This has an effect on the reactivity and thus the function of these related enzymes. In the practical part, methods for the determination of TrxR activity were tested. The specific activity and complete inhibition by methyl viologen of TrxR from Drosophila melanogaster (DmTrxR) and Rattus rattus (rTrxR) were determined using a spectrophotometric method with DTNB substrate. However, the activity of these enzymes could not be determined fluorometrically using fluorescently labeled insulin (FITC insulin) as a substrate, probably due to unsuitable preparation of FITC-insulin. Furthermore, the method for detection of TrxR and GR activity was optimized by specific staining of gels after native PAGE, whereas this detection method seems to be unsuitable for GR. Finally, the expression of TrxR from Apis mellifera was monitored using a qPCR method. A significant decrease in AmTrxR expression was detected in bees fed lipopolysaccharide at 30 and 60 g?ml 1 , which could be the result of a controlled increase in ROS levels in the gut to fight pathogens. However, further measurements are needed to understand the effect of stress factors on TrxR function.
