Study on interaction of mitoxantrone and rhein to selected DNA structures

Abstract:

V této bakalářské práci se věnuji interakci antrachinonových derivátů mitoxantronu a rheinu s vybranými sktrukturami DNA. Mezi zkoumanými strukturami byly jednořetězcová a dvouřetězcová DNA, G-kvadruplex a i-motiv. Pomocí metody cirkulárního dichroismu byly naměřeny teploty tání jednotlivých oligonukleotidů a ty byly porovnány s výsledky naměřeného komplexu oligonukleotid:ligand. Byla také provedena metoda kompetitivní dialýzy pro stanovení afinity studované látky na jednotlivé struktury oligonukleotidů. V závislosti na výsledcích bylo stanoveno, že mitoxantron má významný vliv na stabilitu G-kvadruplexů.

Abstract:

In this thesis I focus on interaction of anthraquinone derivatives mitoxantrone and rhein with selected DNA structures. Single-stranded, double-stranded, G-quadruplex and i-motif DNA structures were studied. Melting points of oligonucleotides (DNA) were measured by circular dichroism and compared with results of oligonucoeotide:ligand complex measurements. Competitive dialysis was performed to determine affinity of studied ligand to certain structures of oligonucleotides. According to results it was determined that mitoxantrone has significant effect on stability of G-quadruplexes.
