Abstract:

Abstrakt Tato disertační práce je prezentována jako kolekce čtyř prací: (1) První práce se zabývá generováním clusterů oxidu wolframu (WxOy+/-) v plynné fázi z pevného WO3 při použití LDI a MALDI time-of-flight hmotnostní spektrometrie s kvadrupólovou iontovou pastí (QIT-TOF-MS). Bylo uvedeno srovnání metod LDI a MALDI s použitím uhlíkových matric. Pro metodu MALDI byly použity nekonvekční uhlíkové matrice jako fulleren, oxidy grafenu a nanodiamanty. Při použití nanodiamantů a oxidů grafenu jako matric byla získána „bohatá“ spektra s ohledem na těžké clustery. (2) Tato studie popisuje identifikaci clusterů přítomných v plazmě generované při interakci laserových pulzů s objemovými vzorky chalkogenidů na bázi arzénu As2Ch3 (Ch= S, Se, Te). Bylo pozorováno, že clustery byly generovány buďto hlubokou fragmentací původní skelné struktury nebo byly formovány během reakce vysoce energetických částic v plazmovém oblouku. Byl vyvinut nový postup pro snižování fragmentace původní skelné struktury za použití „ochranných“ polymerů. Touto metodou bylo generováno velké množství nových těžších clusterů. (3) Třetí kapitola popisuje syntézu magnetických nanokompozitů kobalt-zlato. Syntetizované nanokompozity byly použity jako prekurzor pro generaci ComAun+/− clusterů pomocí LDI TOF hmotnostní spektrometrie a následně byly zkoumány jako aditiva pro analýzu standardních směsí peptidů pomocí MALDI TOFMS. Bylo zjištěno, že nanokompozity zvyšují ionizaci peptidů. Tyto materiály mohou být použity pro detekci dalších biomolekul, a/nebo prekoncentraci analytů s např. jejich následnou magnetickou separací. (4) Poslední kapitola je souhrn věnován etiologii Alzheimerovy choroby, novým metodám diagnostiky a terapie, a dále roli nanočástic.

Abstract:

Abstract The Thesis is presented as a collection of four chapters: (1) The first one describes the generation of tungsten oxide (WxOy+/-) clusters in gas-phase from solid WO3 on using LDI and MALDI with quadrupole ion trap time-of-flight mass spectrometry (QIT-TOF-MS). Comparison of the LDI method with MALDI using non-conventional carbonaceous matrices has been done. For the MALDI method, non-conventional carbonaceous matrices such as fullerene, graphene oxides and nano-diamonds were examined. The ‘rich’ spectra with respect to high mass clusters were obtained when nano-diamonds or graphene oxides were used. (2) The second chapter deals with the identification of the clusters formed in the plasma generated due to the interaction of laser pulses with arsenic chalcogenides As2Ch3 (Ch= S, Se, Te) in bulk forms. It was observed that the clusters were generated either due to the deep fragmentation of original glass structure or they were formed through the reaction of highly energetic species present in the plasma plume. A novel approach has been developed to diminish the deeper fragmentation of the original glass structure by using “protecting” polymers. Using this method many new higher mass clusters were generated. Order of publishing (3) In the third chapter, the synthesis of magnetic cobalt-gold nanocomposites is described. The synthesized nanocomposites were used as a precursor for the generation of ComAun+/- clusters via LDI TOF mass spectrometry and furthermore, these were examined as additives for the analysis of standard peptide mixtures via MALDI TOFMS and found to increase ionization of peptides. This material can be useful also for the detection of other biomolecules, and/or for the preconcentration of analytes and eventually their magnetic separation, for example. (4) The last chapter is review dealing with Alzheimer’s disease etiology, novel diagnosis approaches and therapy, the role of nanoparticles.
