Analysis of interactions of the SMC5/6 complex subunits

Abstract:

In this diploma thesis we studied SMC5/6 complex, which belongs to the group of eukaryotic SMC (Structural maintenance of chromosomes) complexes. The SMC complexes have role in organizations of the chromatin, DNA repair, stabilization of replication bubble and many others. The SMC5/6 complex is composed of proteins SMC5, SMC6 and NSE1-NSE6 subunits. The SMC protein is folding into its final structure, which contains hinge domain, head ATP binding domain and coiled coil arm. SMC5 and SMC6 interact together with their hinge domains and compose a heterodimer, which is the scafold of the entire SMC5/6 complex. Both head domains have the ability to bind ATP and create a temporary dimer. These head domains are also connected together by NSE4 kleisin subunit . NSE4 is in contact with NSE1 and NSE3 subunits and together they compose NSE1/3/4 subcomplex. SMC5/6 complex is also associated with NSE2 subunit (formerly known as Mms21) and NSE5-NSE6 dimer. This thesis deals with characterization of human SMC5/6 protein-protein interactions, more precisely SMC5 protein binding to NSE2, NSE5 and NSE6. For this analysis we created fragments of SMC5 with variable length. After the cloning of these constructs into pGADT7 and pGBKT7 vectors, they were used in Y2H test. In this thesis we were able to identify binding regions of SMC5 with other tested elements and characterized architecture of the human SMC5/6 complex.

Abstract:

V této diplomové práci se věnujeme komplexu SMC5/6, který patří mezi eukaryotické SMC (Structural maintenance of chromosomes) komplexy. SMC komplexy se účastní organizaci chromatinu, oprav DNA, stabilizace replikačních vidlic a mnoha dalších dějů. SMC5/6 se skládá z proteinů SMC5, SMC6 a podjednotek NSE1 až NSE6. SMC proteiny mají zajímavou strukturu, která obsahuje pántovou doménu, hlavovou ATPásove aktivní doménu, a oblast coiled coil ramena. SMC5 a SMC6 spolu interagují přes své pantové domény a tvoří tím heterodimer, který je základem celého SMC5/6 komplexu. Hlavové domény po navázaní dvou molekul ATP pak dimerizují. Tyto hlavové domény jsou propojeny NSE4 kleisínem. NSE4 je asociován s NSE1 a NSE3 a společně tvoří NSE1/3/4 subkomplex. Kromě toho je SMC5/6 komplex asociován s podjednotkou NSE2 (dříve označena jako Mms21) a NSE5-NSE6 dimerem. V rámci této práce se věnujeme charakterizaci interakčních rozhraní lidského SMC5/6 komplexu, konkrétně interakčním oblastem SMC5 s NSE2, NSE5 a NSE6. K analýze protein-proteinových interakcí byly vytvořeny fragmenty SMC5 různých délek. Po jejich klonování do vektorů pGADT7 a pGBKT7 byly použity v Y2H testu. V této práci se podařilo blíže identifikovat interakční oblasti jednotlivých testovaných elementů s SMC5 proteinem a popsat architekturu lidského SMC5/6 komplexu.
