Analysis of the novel components of non-canonical Wnt pathway

Abstract:

Wnt signalizace je jedním z klíčových regulačních mechanismů v průběhu embryogeneze. Uplatňuje se rovněž při udržování homeostáze ve tkáních dospělého organismu. Narušení Wnt signálních drah je proto spojeno s vývojovými poruchami a stejně tak s kancerogenezí. Wnt signalizace se dělí do dvou základních větví: do tzv. kanonické signalizace a signálních drah nekanonických. Zatímco kanonická signalizace, známá také jako Wnt/β-kateninová dráha, spouští prostřednictvím stabilizace β-kateninu transkripci řízenou TCF/LEF, nekanonické dráhy nejčastěji ovlivňují přestavby cytoskeletu. Jedním z receptorů, které se účastní nekanonických Wnt signalizací, je receptorová tyrosin kináza Ror2. Cílem této práce bylo prověřit funkci nové potenciální komponenty Wnt/Ror2 signální dráhy - Nogginu. Tento "cystine knot" protein je znám jako antagonista BMP signalizace. Výsledky prezentované v této práci poskytují důkaz, že Noggin posiluje fosforylaci Dishevelled navozenou stimulací ligandem Wnt5a a že tento jev je závislý na přítomnosti Ror2. Na základě získaných dat byl navržen model interakce Nogginu a Ror2 jak na strukturní, tak i funkční úrovni.

Abstract:

Wnt signalling represents one of the key regulatory mechanisms during embryogenesis. It is also implicated in the maintenance of tissue homeostasis in adult organisms. Thus, disruptions of Wnt pathways are connected with developmental defects as well as with cancerogenesis. Wnt signalling is divided into two main branches: canonical and non-canonical pathways. While the canonical pathway, also known as Wnt/β-catenin pathway, triggers TCF/LEF-regulated transcription via stabilization of β-catenin, noncanonical signalling mostly influences cytoskeleton reorganization. One of the receptors involved in the non-canonical Wnt pathways is the receptor tyrosin kinase Ror2. The aim of this work was to examine the function of a new putative Wnt/Ror2 pathways component - Noggin. This cystine knot protein is known to be a BMP signalling pathway antagonist. The results presented in this thesis are providing an evidence that Noggin enhances Wnt5a-driven Dishevelled phosphorylation in Ror2-dependent manner. Based on the obtained data a model of the interaction between Noggin and Ror2 on both structural and functional levels is proposed.
