Abstract:

Práce se zabývá studiem vztahu buněčné viability a koncentrace glutathionu po působení toxinů s různým mechanismem účinku. U buněčných linií A549 a Jurkat byl studován účinek CDDP, DEM, DMSO a tBHP. Ke studii byl využit WST-1 test pro sledování životaschopnosti buněk po expozici toxinům. Hladina intracelulárního glutathionu byla stanovena pomocí fluorescenční sondy monochlorobiman s fluorometrickou detekcí. Buněčné linie A549 a Jurkat byly inkubovány 1, 3, 6, 18, 21 a 24 hodin s vybranými toxiny o různé koncentraci. Bylo zjištěno, že u buněčné linie Jurkat dochází k poklesu viability před poklesem koncentrace intracelulárního glutathionu po expozici DMSO a tBHP. U působení toxinu DEM na buňky Jurkat a ovlivnění buněk A549 všemi vybranými toxiny předchází pokles GSH poklesu buněčné viability. Z celkových výsledků vyplývá, že buněčná linie A549 je k působení námi vybraných toxických látek méně citlivá než buněčná linie Jurkat.

Abstract:

The thesis deals with studying cell viability and glutathione concentration after treatments with toxins of different mechanism of action. CDDP, DEM, DMSO a tBHP treatments were studied on A549 and Jurkat cell lines. The WST-1 test was used for monitoring cell viability after toxin exposure in this study. A level of intracellular glutathione was determined by a fluorescence probe monochlorobimane followed by fluorometric detection. The A549 and Jurkat cell lines were incubated for 1, 3, 6, 18, 21 and 24 hours with selected toxins at various concentrations. We found that a decrease of cell viability is preceded by depletion of intracellular glutathione after exposure to DMSO and tBHP in Jurkat cells. The reduction of GSH levels is preceded by reduction of cell viability after exposition of DEM to Jurkat cells and after treatment of A549 cells with all selected toxins. In conclusion, the A549 cells are less sensitive to the acting of selected toxins than the Jurkat cells
