Structure and variability of the major histocompatibility complex genes in the Grey Partridge and their effect on fitness

Abstract:

Geny hlavního histokompatibilního komplexu (MHC geny) tvoří nezbytnou část specifické imunity čelistnatých obratlovců. Kódují transmembránové glykoproteiny, které jsou společně s T lymfocyty zodpovědné za rozeznání cizorodých antigenů a spuštění adaptivní imunitní reakce. Struktura MHC genů se výrazně liší u různých skupin obratlovců, stejně jako mechanismus jejich působení na fitness jedince. Velké mezidruhové rozdíly byly nalezeny obzvláště u ptáků. Cílem této práce bylo (1) zoptimalizovat metodu analýzy polymorfní části MHC genu třídy IIB u koroptve polní (Perdix perdix), (2) popsat strukturu této MHC oblasti, individuální a populační variabilitu a pokusit se najít důkazy historické selekce působící na tyto geny a (3) provést analýzy případných vztahů MHC genotypu a fitness jedince ve volně žijící a chovné populaci. Koroptev polní má nejméně dva lokusy MHC genu třídy IIB (jednotliví jedinci vykazovali 2 až 4 různé sekvence exonu 2, tj. části genu, který kóduje oblast MHC glykoproteinu zodpovědnou za rozeznání antigenu), které jsou oba pravděpodobně funkční. Dále bylo prokázáno, že na MHC IIB geny působí (nebo působila) silná pozitivní selekce a v exonu 2 byly nalezeny také nejméně dvě místa rekombinace. Souvislost variability MHC genů a fitness byla nalezena u populace chované v zajetí (např. MHC byly potvrzeny jako tzv. komplementární geny, které signifikantně ovlivňují líhnivost vajec), avšak jejich role v přírodní populaci je zatím nejasná a vyžaduje detailnější studium. Získané poznatky přináší prvotní informaci o MHC regionu u koroptve polní a přispívají tak k pochopení evoluce imunitních genů u ptáků, obzvláště u hrabavých.

Abstract:

Major histocompatibility complex (MHC) genes form an essential part of adaptive immune system in jawed vertebrates. They encode transmembrane glycoproteins, which are together with T lymphocytes responsible for recognition of non-self antigens and initiation of the adaptive immune response. Structure of MHC genes is very variable among vertebrate species, as well as the mechanisms of their effects on individual fitness. Large differences can be found especially among bird species. Aims of this study were (1) to optimise the methods of analysis of the polymorphic part of MHC class IIB gene in the Grey Partridge (Perdix perdix), (2) to describe its structure, individual and population variability and to detect signs of historical selection that have acted on these genes and (3) to analyse possible associations of MHC genotype with individual fitness in the free-ranging and captive populations. The Grey Partridge display at least two loci of MHC class IIB gene (i.e. individuals expressed between 2 and 4 sequence variants of exon 2 encoding part of the peptide-binding region in MHC glycoprotein), which are both presumably functional. The results provided evidence for strong positive selection acting mainly on peptide-binding sites and at least two recombinant points were found in the sequence of exon 2. Evidence for association of MHC genes and fitness was found in the captive population (e.g. MHC were confirmed as "complementary genes" significantly affecting hatchability of eggs), but their role in the free-ranging population is ambiguous, hence requiring further study. The data gained within this study provide the first information about MHC region in the Grey Partridge and contribute to better understanding of evolution of immune genes in birds, especially galliforms.
