Decarboxylation activity of the bacteria Serratia marcescens

Abstract:

Diplomová práce se zabývá sledováním dekarboxylázové aktivity bakterie Serratia marcescens a její produkce biogenních aminů. V první části jsou charakterizovány biogenní aminy se zaměřením na jejich fyziologické účinky, tvorbu a výskyt v potravinách. Další část je zaměřena na popis enzymů a to hlavně na skupinu dekarboxyláz a faktory ovlivňující jejich činnost. Poslední pasáž je věnována enzymové aktivaci a inhibici. V praktické části byly aplikovány vybrané faktory ovlivňující růst bakterie a její dekarboxylázovou činnost. Produkce biogenních aminů byla stanovena pomocí iontově výměnné chromatografie s postkolonovou derivatizací a spektrofotometrickou detekcí. Optimální pH pro růst Serratie marcescens bylo pH 7 {--} 8, zatímco dekarboxylázy projevily největší aktivitu při pH 6. Monosacharidy stimulovaly růst bakterie a zvyšovaly tvorbu putrescinu při pH 6. Naopak, při pH 7 a 8 došlo vlivem přídavku monosacharidů k inhibici produkce putrescinu.

Abstract:

Diploma thesis deals with monitoring decarboxylase activity of the bacteria Serratia marcescens and its production of biogenic amines. In the first part, there is described characteristic of biogenic amines focused on his physiological effects, formation and occurence in foods. The next part is aimed on the description of enzymes, especially group of decarboxylases and the factors that influence their activity. The last passage is applied to enzymic activation and inhibition. In the practical part, there were applied the chosen factors affecting growth of the bacteria and its decarboxylase activity. The production of biogenic amines was assessed with use of ion-exchange chromatography with post-column ninhydrine derivatization and spectrophotometric detection. The optimal pH for S. marcescens growth was 7 {--} 8, while decarboxylases exhibited their maximum activity at pH 6. The monosaccharides stimulated growth of bacteria and increased the production of putrescine at pH 6. On the contrary addition of monosaccharides at pH 7 and 8 led to inhibition in production of putrescine.
