Hepatotoxické a hepatokarcinogenní účinky chemických látek v kulturách buněčných sféroidů

Hepatotoxic and hepatocarcinogenic effects of chemicals in spheroid cell cultures

Abstract:

Mnoho studií poukazuje na důležitou roli jaterních progenitorových buněk, resp. oválných buněk, v hepatokarcinogenezi. V progenitorových buňkách může při poškození jater v důsledku onemocnění nebo působení chemických látek docházet k poškození genetické informace, ovlivnění jejich proliferace, přežívání a schopnosti diferencovat do různých typů buněk. Právě narušení těchto procesů v progenitorových buňkách může souviset se vznikem nádorů. Důležitým nástrojem pro in vitro studium toxických, karcinogenních i terapeutických účinků chemických látek jsou tzv. mnohobuněčné sféroidy. Sféroidy jsou malé agregáty adherentních buněk kultivované bez přichycení k podpůrným materiálům, kdy dochází k maximálním interakcím mezi buňkami a jimi produkovanou extracelulární matrix, a k napodobení reálné situace v tkáni. V této práci byly k vytvoření sféroidů použity buněčné linie WB-ras (tumorigenní) a WB-neo (normální) odvozené od linie WB-F344, která vykazuje vlastnosti jaterních progenitorových (oválných) buněk. Výsledky ukázaly, že buňky tumorigenní linie tvoří stabilní kompaktní sféroidy s hladkým okrajem, oproti normální linii, která vytváří nepravidelné volné agregáty, které se postupně rozpadají. Na sféroidech WB-ras a WB-neo byly hodnoceny účinky vybraných látek (5-fluorouracilu, azidu sodného, microcystinu-LR, cylindrospermopsinu a tetradekanoyl forbol acetátu). Látky s toxickým, cytostatickým nebo nádorově promočním účinkem působily v kulturách sféroidů rozdílně (rozpad a úhyn sféroidů, zpomalení nebo stimulace růstu) a rozdílné bylo i jejich působení na sféroidy normálních vs. tumorigenních buněk (selektivní stimulace růstu tumorigenních buněk působením nádorových promotorů).

Abstract:

Many studies indicated the importance of liver progenitor cells, respectively oval cells, in liver toxicity and hepatocarcinogenesis. During liver injury or chemical exposures, alterations of genetic information, cell proliferation, survival or differentiatoin can occur in the progenitor cells and contribute to various liver diseases. Disruptions of progenitor cell behavior have been linked to the liver carcinogenesis. Multicellular spheroids have become very important tool for in vitro research of toxic, carcinogenic and therapeutic effects of chemicals. Spheroids are a small aggregates of adherent cells cultured without an attachment to the surrounding surfaces or scaffolds, which stimulates interactions between cells and secreted extracellular matrix and cell-cell interactions, and better mimics conditions occurring in vivo in tissues. In this thesis, spheroids of the WB-ras (tumorigenic) and WB-neo (normal) cell lines were used. Both cell lines were derived from WB-F344 cells, which have many characteristics of liver progenitor (oval) cells. The results showed that tumorigenic cells form stable and compact spheroids with a smooth surface. In contrast to tumorigenic line, the normal one formed irregularly shaped loose aggregates, which were shrinking and loosening during the cultivation. Effects of selected chemicals (5-fluorouracil, sodium azide, microcystin-LR, cylindrospermopsin and tetradecanoyl phorbol acetate) on the growth and viability of WB-ras and WB-neo spheroids were evaluated. Chemicals elicited toxic, cytostatic or tumor promoting effects characterized by the spheroid decay and viability reduction, inhibition or stimulation of spheroid growth. There were differences between their effects in the normal and tumorigenic cells, such as selective growth stimulation of tumorigenic cells caused by tumor promoters.
