Ciliogenesis regulation

Abstract:

Cilum je pohyblivá či nepohyblivá řasinka na povrchu buňky, kterou je vybavena většina eukaryot. V posledních dvou dekádách bylo prokázáno, že funkce cilia spočívá kromě lokomoce buňky také v příjmu extracelulárních podnětů, které mohou řídit například vývin či homeostázu živočichů. Stavba a fungování cilia jsou řízeny komplexními buněčnými mechanismy, jejichž dysfunkce vede k onemocněním nazývaným ciliopatie. Tato práce se zabývá procesem vzniku cilia a jeho regulací. Jejím hlavním cílem je popsat distální výběžky mateřské centrioly z hlediska proteinového složení a funkce a uvést je jako strukturu nezbytnou pro ciliogenezi.

Abstract:

Cilium is a motile or immotile organelle on the surface of most eukaryotic cells. In the last two decades it has become clear that cilium is important not only for the locomotion of the cell, but also for the cell’s ability to sense extracellular cues, which may regulate ontogenesis or homeostasis in animals. Ciliary function and ciliogenesis are complex processes which are tightly regulated by the cell and their disruption leads to a group of diseases called ciliopathies. This thesis focuses on ciliogenesis and it’s regulation mechanisms. The main goal of this thesis is to analyze distal appendages of the mother centriole considering their molecular composition and function and to present them as a structure critical for ciliogenesis.
