Effect of acclimation irradiance on the photosynthetic characteristics of Norway spruce

Abstract:

Diplomová práce se věnuje problematice aklimace fotosyntetického aparátu vyšších rostlin na zvýšenou ozářenost. Teoretická část shrnuje problematiku nefotochemického zhášení, jeho vznik, dělení, vlivy a důsledky pro fotosyntetický aparát a jeho přizpůsobení se zvýšené ozářenosti. Cílem experimentální práce bylo zjistit, jak dokáže fotosyntetický aparát smrku (Picea abies L. Karst.) o stáří 27-30 dní aklimovaný na různou intenzitu fotosynteticky aktivní radiace (20, 100, 800 ?mol m-2 s-1) reagovat na dané intenzity aktinického světla, resp. teploty při měření fluorescence chlorofylu a jaký má aklimace vliv na obsah pigmentů violaxantinového cyklu. Výsledky experimentů ukázaly, že se zvyšující se intenzitou aktinického světla klesala schopnost vzorků smrku deexcitovat fotochemickou cestou a navyšovala se míra nefotochemického zhášení. Rostliny aklimované při vyšší ozářenosti byly také schopny lépe využívat fotosyntetickou cestu při vyšších intenzitách aktinického světla. Dále bylo potvrzeno, že smrk patří mezi rostliny relativně odolné vůči zvýšeným teplotám. Při zjišťování obsahu pigmentů bylo potvrzeno, že s narůstající aklimační ozářeností roste po měření fluorescence chlorofylu obsah deepoxidovaných pigmentů xantofylového cyklu, a že smrk obsahuje zeaxantin nekonvertibilní ve tmě, a proto pro tuto rostlinu není vhodné pro výpočet míry nefotochemického zhášení využívat tzv. Stern-Volmerova zhášení maximální fluorescence.

Abstract:

The diploma thesis deals with acclimation of photosynthetic apparatus of higher plants to increased irradiance. The theoretical part summarizes the area of non-photochemical quenching, its formation, classification, effects and consequences for the photosynthetic apparatus and its acclimation to increased irradiance. The goal of the experimental work was to find out how the photosynthetic apparatus of the Norway spruce (Picea abies L. Karst.), 27 30 days old, acclimated to various intensities of photosynthetically active radiation (20, 100, 800 ?mol m-2 s-1), can react to specific intensity of actinic light or temperature during chlorophyll fluorescence measurement, and what is the impact of acclimation on the amount of the violaxanthin cycle pigments. The results of experiments showed that with increasing intensity of actinic light the ability of the spruce samples to de-excite through the photochemical pathway decreased and the rate of non-photochemical quenching increased. Plants acclimated to higher irradiance were also able to use the photosynthetic pathway better at higher intensities of actinic light. It was also confirmed that spruce ranks amongst plants relatively resistant to increased temperatures. Measurements of the pigments amount confirmed that with the increasing acclimation irradiance the amount of de-epoxidized pigments of the violaxanthin cycle increases after fluorescence measurements and that the spruce contains zeaxanthin nonconvertible in the dark, therefore for spruce it is not suitable to use the Stern-Volmer maximum fluorescence quenching method to calculate the rate of non-photochemical quenching.
