Haplogenotype analysis in mannose-binding lectin gene and dental caries in children

Abstract:

Zubní kaz je multifaktoriální onemocnění s vysokou prevalencí u dětské populace. Důležitou roli v etiopatogenezi tohoto onemocnění má i stav orální mikrobioty, který může být ovlivněn koncentrací lektinu vázajícího manózu (MBL). MBL je schopen rozpoznávat sacharidové struktury na povrchu různých mikroorganizmů a prostřednictvím aktivace lektinové dráhy komplementu uvést v činnost imunitní systém. Deficit MBL je způsoben variabilitou v genu MBL2, zejména kombinací šesti funkčních jednonukleotidových polymorfizmů (SNPs). Cílem této práce bylo provést optimalizaci metody SNaPshot pro genotypizaci výše zmíněných SNPs v genu MBL2 a následně v designu asociační studie kontrol a případů stanovit tyto genové varianty u vzorků DNA získaných od dětí trpících zubním kazem a dětí s intaktním chrupem. Výsledkem této práce je optimalizovaná metodika SNaPshot a v rámci genetické asociační studie byly spojeny SNPs MBL2 rs5030737 a rs7095891 se vznikem a rozvojem zubního kazu (p < 0,05). Jedná se o první studii, která zkoumala asociaci všech šesti významných SNPs v genu MBL2 se zubním kazem.

Abstract:

Dental caries is a multifactorial disease with a high prevalence in the paediatric population. Oral microbiota plays an important role in the etiopathogenesis of this disease and may be affected by levels of mannose binding lectin (MBL). MBL can recognize carbohydrate structures on the surface of various microorganisms and set the immune system in motion by activating the lectin pathway of the complement. MBL deficiency is caused by the variability in the MBL2 gene, particularly by a combination of six functional single nucleotide polymorphisms (SNPs). The aim of this thesis was to optimize SNaPshot assay for genotyping the above-mentioned SNPs in the MBL2 gene and subsequently to determine these gene variants in DNA samples, obtained from children with dental caries and healthy children, in the design of a case-control association study. The result of this thesis is an optimized SNaPshot assay and association of MBL2 SNPs rs5030737 and rs7095891 with dental caries (p < 0,05). This is the first study to examine all six important SNPs in the MBL2 gene in association with dental caries.
