Potential modulation of cancer stem cell phenotype in neuroblastoma

Abstract:

Nádorové kmenové buňky (CSCs) jsou vysoce agresivní nediferencované buňky, jimž je přisuzována klíčová role při nádorovém bujení. Zároveň se velkou měrou podílejí na rezistenci tumorů vůči konvenčním terapiím, vzniku metastáz a relapsů onemocnění. Přítomnost CSCs byla potvrzena u mnoha typů nádorů, včetně neuroblastomu. Neuroblastom patří k nejčastějším solidním nádorům dětského věku. Jedná se o vysoce heterogenní onemocnění s širokým rozsahem klinického průběhu, od spontánně regredujících nádorů až po vysoce agresivní. U nejvíce rizikových neuroblastomů se jako jeden z možných důvodů velké agresivity, rezistence a relapsů těchto nádorů uvádí právě přítomnost CSCs. Pro zlepšení léčby bývá proto u těchto stádií neuroblastomu po chemoterapii a transplantaci hematopoetických kmenových buněk zařazen krok indukované diferenciace vyvolané podáním retinoidů. Tyto deriváty vitaminu A jsou známými diferenciačními činidly a předpokládá se jejich účinek při udržování minimální reziduální choroby a prevenci relapsu onemocnění. Dosud ale nebylo jasně prokázáno, zda retinoidy skutečně vyvolávají diferenciaci neuroblastomových CSCs. Tato práce se věnuje analýze změn vyvolaných působením all-trans retinové kyseliny (ATRA) u tří pacientských neuroblastomových buněčných linií a referenční linie SH-SY5Y. Při charakterizaci buněčných linií byla zjištěna lokalizace vybraných markerů CSCs a status genu MYCN. V další části práce byl sledován vliv ATRA na diferenciaci, kmenovost a tumorigenicitu neuroblastomových buněk. Zatímco diferenciační potenciál ATRA byl potvrzen za použití pouze části metod, určité snížení kmenovosti bylo v důsledku ovlivnění retinoidy pozorováno u buněk všech sledovaných linií.

Abstract:

Cancer stem cells (CSCs) are highly aggressive undifferentiated cells which are implicated to play a key role in tumorigenesis. At the same time, they are involved in the resistance of tumors, metastasis and relapse of the disease. CSCs have been reported in many types of tumors, including neuroblastoma. Neuroblastoma is one of the most common solid tumors in children. It is a highly heterogenous disease with a widely variable clinical course, ranging from spontaneously regressing tumors to highly aggressive cases. A presence of the CSCs is reported to be one of the possible reasons for high aggressiveness, resistance and relapses of high-risk neuroblastomas. Therefore, an induced differentiation by retinoids is included in the therapy of these high-risk tumors following the chemotherapy and hematopoietic stem cell rescue to improve the treatment. These Vitamin A derivates are well-known differentiating agents and expected to maintain minimal residual disease and to prevent tumor relapse. However, it has not yet been clearly demonstrated whether retinoids actually induce differentiation of neuroblastoma CSCs. This thesis analyzes all-trans retinoic acid (ATRA)-induced changes in three patient-derived neuroblastoma cell lines and established cell line SH-SY5Y. During the characterization of the used cell lines, the localization of the selected CSCs marker and the MYCN gene status were determined. In the following part of the work, the impact of ATRA on the differentiation, stemness and tumorigenicity of neuroblastoma cells was monitored. While the differentiation potential of ATRA was confirmed using only some methods, certain decrease in stemness caused by retinoids was observed in the cells of all studied cell lines.
